이중 가닥 DNA의 상보적 가닥들은 서로 다른 속도로 복제됩니다. 한쪽 가닥에서는 복제 과정이 연속적이고 빠르게 진행되며, 이렇게 새로 형성된 딸가닥을 선도 가닥이라고 합니다.
다른 가닥의 경우, 복제 과정은 불연속적이고 상대적으로 더 느리며 약간 더 늦게 시작됩니다. 이 딸가닥을 지연 가닥이라고 합니다.
DNA polymerase는 5'에서 3' 방향으로만 DNA를 합성할 수 있습니다. 이로 인해 선도 가닥은 연속적으로 합성됩니다.
하지만 DNA polymerase는 지연 가닥(lagging strand)에서 3'에서 5' 방향으로 DNA를 합성할 수 없습니다.
