Membrane Carbohydrates

탄수화물은 지질과 단백질에 이어 생체막의 세 번째 주요 구성 요소입니다.
탄수화물이 결합된 단백질인 당단백질(Glycoprotein)은 막 탄수화물의 거의 90%를 차지합니다. 당지질은 나머지 10%입니다.
당단백질(glycoprotein)과 당지질(glycolipid)은 모두 막의 바깥쪽에서 발견되며 항원 및 수용체 역할을 포함하여 다양한 기능을 수행합니다.
탄수화물은 N-글리코시드 및 O-글리코시드 결합을 통해 단백질에 부착됩니다.
N-글리코시드 결합은 아스파라긴의 아미드기와 N-아세틸글루코사민과 같은 당 사이에서 발생하는 반면, O-글리코시드 결합은 세린 또는 트레오닌의 수산기와 N-아세틸갈락토사민과 같은 당 사이에 형성됩니다.
결합 탄수화물은 단백질 분해 효소로부터 단백질을 보호하고, 전체 충전량을 변화시키며, 단백질의 수분 결합 능력을 증가시킵니다.
당지질은 스핑고지질(sphingolipid)인 세라마이드(ceramide)에서 파생됩니다. 글루코세레브로시드 또는 갈락토세레브로사이드는 포도당 또는 갈락토오스가 세라마이드에 결합할 때 합성됩니다.

막 탄수화물

원형질막은 지질, 단백질, 탄수화물로 구성된 동적 장벽입니다. 이는 세포 성장과 생존에 필요한 많은 세포 과정의 진원지입니다. 탄수화물은 원형질막이 기능을 효과적으로 수행하는 데 도움이 되는 독특한 구조적, 화학적 특성을 가지고 있습니다.

막 탄수화물은 소수성 영역이 없으며 세포의 외부 표면에만 위치합니다. 당 분자의 추가 또는 단백질의 글리코실화는 소포체와 골지체의 내강에서 일어나는 반면, 지질의 글리코실화는 골지체의 내강에서만 발생합니다. 따라서 소포가 골지체에서 싹이 트면 당지질과 당단백질이 내부 층에 존재합니다. 그러나 소포가 원형질막과 융합되면 내부 층과 관련 당지질 및 당단백질이 외질 측면에 노출됩니다. 당단백질과 당지질이 바깥층에서만 발견되는 이유입니다. 일부 진핵세포와 박테리아 세포는 세포 외부로 다량의 탄수화물을 분비하여 세포 주위에 당질막으로 알려진 설탕 코팅을 형성합니다.

탄수화물은 단백질과 DNA보다 구조적으로 더 다양합니다. 단백질과 DNA에서는 주로 펩타이드와 포스포디에스테르 공유결합이 각각 관찰된다. 대조적으로, 각 설탕 분자는 여러 유형의 공유 결합을 형성할 수 있으며 동일한 설탕 분자에서 서로 다른 올리고당 구조를 형성할 수 있습니다. 탄수화물의 구조적 다양성은 각 세포 유형에 뚜렷한 정체성을 부여하는 데 도움이 되며 다양한 세포 기능에 필수적인 역할을 합니다. 예를 들어, 인간의 적혈구(RBC) 표면에 존재하는 항원에 따라 혈액형은 -A, B, AB, O의 네 가지 유형으로 분류됩니다. Membrane carbohydrates help give cells their surface identity and support plasma membrane function. The plasma membrane is a dynamic barrier made of lipids, proteins, and carbohydrates. These sugar chains have unique structural and chemical traits that help the membrane work properly for cell growth and survival.

Membrane carbohydrates do not have a hydrophobic region, so they are found only on the outer surface of the cell. Sugar addition, or glycosylation, of proteins occurs in the lumens of the endoplasmic reticulum and the Golgi apparatus. Glycosylation of lipids occurs only in the lumen of the Golgi apparatus.

When vesicles bud from the Golgi apparatus, glycolipids and glycoproteins are on the inner layer. After these vesicles fuse with the plasma membrane, that inner layer faces the outside of the cell. This is why glycolipids and glycoproteins are found only on the exoplasmic side.

Some eukaryotic and bacterial cells also release large amounts of carbohydrate outside the cell. This forms a sugar coat called the glycocalyx. Carbohydrates are more structurally diverse than proteins and DNA because each sugar molecule can form multiple kinds of covalent bonds. That allows many different oligosaccharide structures to form from the same sugars.

This diversity helps each cell type have a distinct identity and supports many cellular functions. A clear example is human red blood cell antigens, which determine blood groups A, B, AB, and O. Type A blood has A antigen and anti-B antibodies, type B has B antigen and anti-A antibodies, type AB has both antigens and no antibodies against them, and type O has neither antigen but carries antibodies to both.
