Dual In Vivo Imaging

Dual in vivo imaging is the use of two complementary imaging approaches to visualize biological structures, processes, or responses in a living organism, improving the information available from a single technique. The methods may acquire signals simultaneously or sequentially and combine measurements through spatial co-registration, pairing anatomical detail with molecular, functional, or cellular readouts from modalities such as optical imaging, ultrasound, or magnetic resonance imaging. In bioengineering, dual in vivo imaging supports noninvasive monitoring of tissue engineering, drug delivery, disease progression, and biomaterial performance, while enabling researchers to relate structure to function over time and reduce the need for repeated invasive sampling.

With its small transparent body, well-documented neuroanatomy and a host of amenable genetic techniques and reagents, C. elegans makes an ideal model organism for in vivo neuronal imaging using relatively simple, low-cost techniques. Here we describe single neuron imaging within intact adult animals using genetically encoded fluorescent calcium indicators.

This video demonstrates the use of dual fluorescence imaging to evaluate the differentiation of oligodendrocytes from precursor cells (OPCs) isolated from a rat pup brain. The process includes fixing the cells, labeling specific markers with primary and fluorophore-conjugated secondary antibodies, and...

The in vivo measurement of smooth muscle contractions along the gastrointestinal tract of laboratory animals remains a powerful, though underutilized, technique. Flexible, dual element strain gages are not commercially available and require fabrication. This protocol describes the construction of reliable, inexpensive strain gages for acute or chronic implantation in rodents.
