Flatbed Scanner Imaging

Flatbed scanner imaging is a contact-based method that converts two-dimensional specimens or objects into digital images, providing a rapid, noninvasive record for biological analysis. A glass platen supports the sample while a moving light source illuminates it; reflected or transmitted light is captured line by line by a charge-coupled device or contact image sensor and translated into pixel values. In biological techniques, scanners can document colony growth, tissue sections, gels, plates, and other relatively flat samples for measurements of area, intensity, morphology, or spatial distribution. Standardized imaging conditions and image-analysis software improve reproducibility, enable quantitative comparisons, and create archival records for research and teaching.

Cited by 5 •

2016

A scanner for imaging magnetic particles in planar samples was developed using the planar frequency mixing magnetic detection technique. The magnetic intermodulation product response from the nonlinear nonhysteretic magnetization of the particles is recorded upon a two-frequency excitation. It can be used to take 2D images of thin biological samples.

Source: Bru, J., et.al. Time-lapse Imaging of Bacterial Swarms and the Collective Stress Response. J. Vis. Exp. (2020)This video demonstrates how to visualize and measure swarm avoidance behavior in Pseudomonas aeruginosa using a swarming agar plate assay. It shows the setup for time-lapse imaging under controlled conditions and explains how antibiotic-induced stress signals redirect bacterial movement, which is quantified through image analysis.
