Protein Lysate Digestion

Protein lysate digestion is a sample-preparation technique that converts the complex mixture of proteins extracted from cells or tissues into smaller peptides for proteomic analysis. Typically, proteases such as trypsin cleave peptide bonds at defined amino acid residues under controlled buffer, temperature, and incubation conditions, producing fragments compatible with liquid chromatography and tandem mass spectrometry. In cancer research, this process supports identification and relative quantification of tumor-associated proteins, signaling molecules, and potential biomarkers across experimental samples. Reliable digestion improves peptide recovery, analytical reproducibility, and the ability to compare molecular changes linked to cancer development, treatment response, and disease progression.

Protein Digestion

Protein digestion begins in the stomach, where the highly acidic environment can easily disrupt protein structure by exposing the peptide bonds of polypeptide chains. After polypeptide chains are broken into individual amino acids by a series of digestive enzymes, the amino acids are transported to the liver via the bloodstream to produce energy. Pepsin Pepsin is a protease, or protein-digesting enzyme, that is produced in the stomach and is one of the main digestive enzymes in the human...
