Half Life Duration Effect

Half-life and duration of effect describe related but distinct features of a drug’s behavior in the body. Half-life is the time required for the drug concentration in plasma to decrease by half, whereas clinical duration depends on how long concentrations remain above the therapeutic threshold and how strongly the drug interacts with its target. Distribution, metabolism, elimination, active metabolites, and receptor binding can therefore make a drug’s effects shorter or longer than its plasma half-life suggests. Understanding this relationship supports dose selection, dosing intervals, therapeutic monitoring, and prediction of drug accumulation or prolonged effects in clinical practice.

Drug elimination from the body primarily occurs through metabolic and excretion pathways. Hepatic metabolism transforms lipophilic drugs into hydrophilic forms for excretion, typically via enzymatic processes classified as phase I (modification) and phase II (conjugation). Renal excretion eliminates drugs and metabolites through filtration and secretion in the kidneys. Impairment in liver or kidney function can hinder these processes, delaying drug clearance and extending the drug’s half-life.

A lengthening in meal duration represents orofacial nociceptive behavior in rodents similar to the guarding behavior of humans with orofacial pain.
