Muscle Wasting

Muscle wasting is the progressive loss of skeletal muscle mass, strength, and function, and it is an important clinical sign of immobility, chronic disease, aging, or inadequate nutrition. It develops when muscle protein breakdown exceeds synthesis, often through reduced mechanical loading, impaired nerve stimulation, inflammation, hormonal changes, or disrupted energy balance. Clinicians assess muscle wasting using physical examination, strength testing, imaging, and measures of body composition to identify underlying causes and track progression. Understanding these mechanisms supports rehabilitation, nutritional and pharmacological interventions, and improved management of conditions such as cancer, chronic infection, neuromuscular disorders, and prolonged hospitalization.

We describe a method to directly measure muscle force, muscle power, contractile kinetics and fatigability of isolated skeletal muscles in an in vitro system using field stimulation. Valuable information on Ca2+ handling properties and contractile machinery of the muscle can be obtained using different stimulating protocols.
