Islet Adhesion Technique

The Islet Adhesion Technique is a laboratory method for securing pancreatic islets to a culture surface, making these three-dimensional cell clusters easier to maintain, image, and analyze. It works by allowing islets to settle onto an adhesive or extracellular-matrix-coated substrate, where cell–matrix interactions promote attachment and reduce movement during handling or microscopy. In developmental biology, this approach supports studies of islet organization, cell differentiation, and maturation under controlled culture conditions. By preserving intact islet architecture while improving experimental access, the technique can facilitate morphological analysis, live-cell observation, and investigations of how cellular interactions influence endocrine tissue development.

Confocal Imaging of Neuropeptide Y-pHluorin: A Technique to Visualize Insulin Granule Exocytosis in Intact Murine and Human Islets

We describe a protocol for visualization of insulin exocytosis in intact islets using pHluorin, a pH-sensitive green fluorescent protein. Isolated islets are infected with adenovirus encoding pHluorin coupled to the vesicle cargo neuropeptide Y. This allows for the detection of insulin granule fusion events by confocal microscopy.

This video describes the technique of imaging islet cells in real-time, to study the changes in cytosolic calcium concentration in the minor subpopulations of the pancreatic islet cells. Minor cell subpopulations can be identified using functional responses to marker compounds, such as adrenaline and ghrelin, that induce changes in cytosolic calcium in a specific populations of islet cells. This can provide an insight into the cross talk between the blood glucose and the islet secretory output.
