Archived Tissue Specimens

Archived tissue specimens are preserved biological samples retained after collection for later examination, providing a record of cellular material and genetic information. When storage conditions limit chemical and enzymatic degradation, researchers can retrieve tissue, isolate nucleic acids, and use amplification or sequencing workflows to examine inherited variants, somatic mutations, gene expression, or genomic changes. In genetics, these specimens support retrospective studies, validation of molecular tests, analysis of disease-associated variation, and comparisons across patients, time points, or populations. Their value depends on specimen quality, preservation history, and the integrity of recovered material, making careful documentation and standardized processing essential for reproducible results.

Some researchers gain access to large amounts of data without interacting with a single research participant. Instead, they use existing records to answer various research questions. This type of research approach is known as archival research. Archival research relies on looking at past records or data sets to look for interesting patterns or relationships. For example, a researcher might access the academic records of all individuals who enrolled in college within the past ten years and...

(2016)This video demonstrates the immunolabelling of freeze-fractured replicas of mouse brain tissue for electron microscopy. The fractured and coated replicas are digested to expose receptors. Primary and gold-tagged secondary antibodies are added to label specific neuronal membrane receptors. The replica...

Using Digital Image Correlation to Characterize Local Strains on Vascular Tissue Specimens

We describe the use of digital image correlation to characterize the local surface strain field on vascular tissue samples subjected to uniaxial tensile testing. These measurements facilitate precise quantification of the sample mechanical response and the generation of constitutive stress-strain relations.

Using Archival Japanese Paper and Thermoplastic Resins to Prepare Fossils for Storage, Display, Transport, and Radiography

2025

This article presents a suite of techniques for reconstructing fossil specimens using paper-resin composites. These composites are made by combining archival-grade paper with thermoplastic resins commonly used in fossil preparation laboratories. The resulting lightweight composites add long-term stability to specimens while retaining suitability for radiological analyses.
