Beta-2 Integrin

Beta-2 integrins are leukocyte-specific cell-surface adhesion receptors that coordinate immune-cell movement, communication, and interactions with other cells and the extracellular environment. Formed by pairing a common CD18 beta subunit with distinct alpha subunits, they bind ligands such as intercellular adhesion molecules and complement fragments; signaling can increase their affinity and promote firm adhesion, spreading, and migration through inflamed tissues. In immunology and infection, beta-2 integrins help neutrophils and lymphocytes exit the bloodstream, recognize targets, and organize antimicrobial responses. Their dysfunction can impair host defense and leukocyte trafficking, while abnormal activation contributes to inflammatory disease, making them important targets for studying immune regulation and therapeutic intervention.

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling." In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.
