Histidine Tag Cleavage

Histidine tag cleavage is the enzymatic removal of a polyhistidine purification tag from a recombinant protein after isolation, yielding a product closer to its native form. The target protein is engineered with a protease recognition sequence between the His tag and the protein, then treated with a site-specific protease such as TEV protease; the cleaved tag and protease can be separated by nickel-affinity chromatography. This process helps reduce effects from the tag on protein folding, activity, or interactions. In immunology and infection research, tag-free antigens, antibodies, and pathogen-derived proteins support structural studies, binding assays, and functional experiments with greater physiological relevance.

We report and demonstrate an optimized nitrocellulose binding assay that can be used to quantify autophosphorylation of purified bacterial histidine kinases. Our method has several advantages over traditional SDS-PAGE based techniques, providing a valuable alternative for characterizing these important proteins.
