Luciferase Substrate

Luciferase substrates are light-producing molecules that react with luciferase enzymes, enabling sensitive detection of biological activity in medical research. In the presence of the appropriate cofactors and conditions, luciferase catalyzes substrate oxidation and generates an electronically excited product that releases measurable photons as it returns to a lower-energy state. Common substrate systems support bioluminescence assays, reporter gene studies, and imaging of cells or molecular processes in living organisms. Because emitted light can be quantified with high sensitivity, luciferase substrates help evaluate gene expression, track disease-related pathways, monitor therapeutic responses, and develop noninvasive diagnostic or drug-screening methods.

Source: Chang, X., et al. Preparation of Pseudo-Typed H5 Avian Influenza Viruses with Calcium Phosphate Transfection Method and Measurement of Antibody Neutralizing Activity. J. Vis. Exp. (2021).This video demonstrates a luciferase-based assay to quantify pseudovirus infection in epithelial cells. The pseudovirus encodes the luciferase gene, which is expressed in infected cells. After washing, and cells are incubated with lysis buffer at an ultra-low temperature, and then thawed to induce lysis.

Source: Chen, Y., et al., Measurement of Specific Mycobacterial Mistranslation Rates with Gain-of-function Reporter Systems. J. Vis. Exp. (2019)This video demonstrates a method to quantify mistranslation in Mycobacterium smegmatis using an inducible GFP and secreted mutant luciferase reporter system. It shows how luminescence resulting from mistranslation is measured and compared to GFP fluorescence to evaluate the impact of a test molecule on translational accuracy.

Luciferase Assay to Study Efficacy of Antiparasitic Chemicals Against Parasites

2025

The video demonstrates the luciferase assay used to determine the efficacy of an antiparasitic chemical inhibitor against the intracellular pathogen Toxoplasma gondii. The study involves luciferase-expressing parasites invading the host cells to replicate and survive. The chemical inhibitor binds with the cysteine proteases of the parasite and inhibits replication. The luciferase activity detected is proportional to the fold changes in parasite growth, determining the drug's efficacy against...

Measurement of the In Vivo Burden of Bacteria Expressing Luciferase in an Insect Larva

This video showcases the in vivo bacterial burden measurement using Bacillus Calmette-Guérin, BCG, lux-infected insect larvae. The process involves larval homogenization, treatment with a luciferase substrate, and bioluminescence measurement to determine relative light units that correlate with bacterial burden.

Luciferase-Based Growth Assay to Quantify Intracellular Parasite Reproduction

2025

In this video, we demonstrate the luciferase assay to quantify the growth of the intracellular pathogen Toxoplasma gondii. Utilizing a strain of Toxoplasma that expresses luciferase enzyme after infecting the host cells, the luciferase activity is detected in the infected cell culture to determine the successful reproduction of the parasite.
