Anesthetized Drosophila Flies

Anesthetized Drosophila flies are fruit flies rendered temporarily immobile and less responsive through controlled anesthesia, providing a practical preparation for neuroscience experiments. Anesthetic exposure, commonly through carbon dioxide or cooling, reduces locomotor activity and sensory responsiveness, allowing researchers to position flies, access neural tissues, or perform precise manipulations while limiting movement-related disturbance. This preparation supports neural circuit imaging, electrophysiological recording, stimulation, and behavioral studies that compare activity during anesthesia with responses after recovery. By standardizing immobilization and recovery conditions, anesthetized Drosophila provide a tractable model for linking brain activity to motor control, sensory processing, and disease mechanisms.

Drosophila Courtship Conditioning: A Method to Test Learning and Memory In Flies

2023

This video describes a classical conditioning assay that tests learning and memory in Drosophila, called courtship conditioning. The assay is based on the reduction of male courtship after experiencing a rejection by a non-receptive premated female. The example protocol shows a setup for the procedure that can be used to assess short- and long-term memory.

Fly Climbing Assay: A Method to Test General Locomotion in Drosophila melanogaster

2023

This video describes a simple method to quantify fly locomotion and shows the climbing assay performed. The fly climbing assay is a general test of motor function which relies on a negative geotaxis response.

An In Vivo Assay to Quantify Bacterial Phagocytosis in Adult Drosophila Flies

2025

This video demonstrates an in vivo assay designed to measure bacterial phagocytosis in adult Drosophila flies. Following the injection of fluorescently labeled bacteria into the abdomen of adult flies, hemocytes associated with the dorsal vessel engage in phagocytosis. The introduction of trypan blue dye serves to quench the fluorescence emitted by extracellular, non-phagocytosed bacteria. This quenching process enhances the identification of phagocytosed bacteria when observed under a...

Drosophila Activity Monitor (DAM): A Method to Measure Locomotor Activity in Flies

2023

This video describes the Drosophila activity monitor (DAM) system used to track locomotor activity. Researchers use activity data collected from the DAM to study circadian rhythms in fruit flies. The featured protocol clip shows how to load flies in the device and record activity data for circadian experiments.
