1. 동물 준비
<ol><li>실험군과 집단당 동물의 수를 미리 결정한다.</li>
	<li>수컷 BALb/c 마우스를 선택하여 22 ± 1 °C의 일정한 온도, 60 ± 5%의 일정한 습도 및 12시간의 일주기 조명 환경에서 보관합니다.</li>
</ol>2. AS 마우스 모델 구축
참고: AS 모델은 고전적인 모델링 방법 16,17,18,19,20,21인 연골 프로테오글리칸(PG)으로 유도되었습니다. BALB/c 균주 마우스를 PG로 반복적으로 면역시켜 건염 및 척추염을 유발했습니다. 이 프로토콜에 사용된 재료, 시약 및 기기와 관련된 자세한 내용은 재료 표를 참조하십시오.
<ol><li>200μL 피펫을 사용하여 200μL의 PG 및 CFA(또는 IFA)를 1.5mL 원심분리 튜브에 넣습니다. 원심분리기 튜브에 있는 액체를 섞기 위해 부드럽게 흔듭니다.</li>
	<li>균질화를 위해 원심분리기 튜브를 조직 분쇄기에 넣은 다음 얼음 위에서 4분 동안 배양합니다. 티슈 그라인더에서 다음 매개변수를 설정합니다: 60Hz, 연삭 30초, 간격 10초 동안 총 6사이클.</li>

</ol>3. 두뜸

	<li>생강 진흙 10g과 생강즙 20mL를 준비하여 나중에 사용할 수 있도록 종이컵에 담습니다.</li>
	<li>TCM 분말(계피 분말, 정향 분말, 노토페리지움 분말, 안타리딘 분말 및 인공 사향의 혼합물)22 0.1g의 무게를 달고 절구에 갈아 넣습니다.</li>
	<li>마우스를 동물 마취 기계의 마취 유도 상자에 넣고 2L/min의 유속으로 2-3% 이소플루란과 산소의 혼합물로 마취합니다.</li>
	<li>초기에 놀람 반사가 없는지 관찰하여 마취 깊이를 평가하고 부드러운 발가락 꼬집기에 반응하여 페달 반사가 없는지 확인하여 수술 마취 수준을 확인합니다.</li>
	<li>마우스가 수술면 마취에 도달한 후 상자에서 꺼내 엎드린 자세로 수술대에 고정합니다. 마우스의 입과 코를 마취제의 배출구에 맞춥니다.</li>
	<li>마취 기계에 부착된 마스크를 통해 마취를 투여하고, 이를 통해 이소플루란과 산소가 비재호흡 회로를 통해 전달됩니다. 마취를 유지하고 전체 수술 절차 동안 마우스를 37°C 가열 패드에 유지하십시오. 건조를 방지하기 위해 눈에 윤활제를 바르십시오.</li>
	<li>경혈을 찾아 자극 부위를 확인합니다(그림 4).</li>
	<li>쥐 뒷면의 살색 피부가 보일 때까지 쥐 뒷면의 털을 면도합니다. 제모 너비가 ~1.5cm이고 길이가 일곱 번째 경추에서 꼬리 척추까지인지 확인합니다(그림 5A).</li>
	<li>면도 부위에 면봉으로 준비한 생강즙을 면도 부위에 바릅니다(그림 3).</li>
	<li>마우스 뒷면의 중앙선에 붓으로 TCM 가루를 골고루 뿌리고 뽕나무 종이로 덮습니다(그림 5B-D).</li>
	<li>생강 진흙으로 길이 6cm, 너비 1.5cm, 두께 3mm의 사다리꼴 생강 기둥을 만듭니다. 뽕나무 종이에 생강 기둥을 고정합니다(그림 5E).</li>
	<li>생강 기둥의 정중선 위에 홈을 만들고(그림 3) 생강 기둥에 뜸쑥 콘을 놓습니다.</li>
	<li>뜸 원뿔에 불을 붙이고 이전 원뿔이 타버린 후 새 뜸 원뿔을 놓습니다. 마우스가 타는 것을 방지하기 위해 생강 기둥의 견고성을 확인하여 총 3개의 원뿔을 태웁니다(그림 5F).</li>
	<li>뜸쑥 콘을 태운 후 생강과 TCM 가루를 닦아내고 마우스 뒷면을 청소합니다. 이 시점에서 마취를 중단하십시오.</li>
	<li>쥐가 음식과 물에 제한 없이 접근할 수 있는 새 케이지에서 의식을 되찾을 수 있도록 합니다. 회복 과정을 돕기 위해 가열된 패드에 케이지를 놓습니다. 동물이 충분한 의식을 회복할 때까지 관찰하십시오.</li>
	<li>마우스가 완전히 회복되면 일반 하우징으로 되돌리기 전에 오른쪽 반사의 존재를 확인하십시오. 알림: 수술 중에는 약한 호흡을 피하기 위해 쥐의 정신 상태를 모니터링해야 합니다. 화상을 입지 않도록 하되 쥐를 따뜻하게 유지하고 발톱과 꼬리에 멍이 들지 않도록 하십시오.</li>
</ol>4. 관절염 지수(AI) 평가
<ol><li>AI 점수를 모델링 전, 모델링 후, 처리 후의 세 번 평가합니다. 평가 기준은 다음과 같습니다 : 0, 발적이나 부종이 없습니다. 1, 몇 개의 발가락에 약간의 발적 또는 부기; 2, 대부분의 발가락 관절과 발가락의 발적과 부기; 3, 발과 발목 관절 아래의 심각한 발적 및 부종; 4, 발과 발목 관절의 발적 및 부종23.</li>
	<li>최대값 16으로 네 발 모두의 AI 점수를 계산합니다.</li>
</ol>5. 로타로드 테스트
알림: 회전 테스트는 마우스가 회전 드럼에서 보내는 시간을 기록하여 운동 조정과 균형을 평가하는 데 사용됩니다. 이 실험은 20분 간격으로 3번의 시행으로 구성되었습니다. 공식 테스트 데이터는 세 번째 실험에서 기록되었으며, 처음 두 번은 훈련 연습으로 사용되었습니다.
<ol><li>마우스를 300초24에 걸쳐 4에서 40rpm까지 연속 가속도로 회전하는 회전 드럼 위에 놓는다.</li>
	<li>대기 시간(마우스가 로드에서 떨어질 때까지의 시간)을 측정합니다. 패딩이 떨어질 때 부상을 방지하기 위해 패딩을 제공하십시오.</li>
</ol>6. 오픈 필드 테스트(OFT)
알림: OFT는 병리학적 행동을 식별하기 위해 자율 운동 상태를 평가하는 데 사용됩니다. 4 개의 정사각형 오픈 필드 경기장 (50cm x 50cm x 40cm)을 함께 배치하여 장치를 형성했습니다. 상자의 바닥은 9개의 동일한 사각형으로 나뉘어져 있습니다.
<ol><li>실험실의 배경 소음을 65dB 미만으로 제어합니다.</li>

</ol>7. 안락사 및 검체 채취

	<li>발가락 꼬집음에 대한 반응으로 페달 반사가 없는지 확인하여 마취 깊이를 확인합니다. 겸자를 사용하여 안구를 잡고 신속하게 제거하여 혈액이 안와에서 미세 원심분리기 튜브로 흐르도록 합니다. 알림: 심장 천자와 같은 대체 혈액 수집 방법도 사용할 수 있습니다.</li>
	<li>혈액 방울 속도가 느려지면 쥐의 심장 부위를 부드럽게 눌러 혈액 펌핑 속도를 높이고 더 많은 혈액을 얻습니다. 그런 다음 자궁 경부 탈구를 사용하여 쥐를 안락사시킵니다.</li>
	<li>주로 사용하지 않는 손의 집게를 사용하여 발목 관절 바로 위의 다리를 잡습니다. 그런 다음 가위를 사용하여 발목 관절 바로 위의 다리를 자릅니다.</li>
</ol>8. 조직학적 분석
<ol><li>발목 표본을 4% 파라포름알데히드에 24시간 이상 담그십시오. 1개월 동안 10% EDTA(pH = 7.4)로 석회질을 제거합니다.</li>

	<li>조각에 에오신을 1분 동안 얼룩지게 하고 증류수로 3분 동안 헹굽니다.</li>
	<li>에탄올 농도가 증가하는 그라데이션(80%, 95%, 100%, 100%)에 각각 1분 동안 슬라이스를 담그십시오. 크실렌에 3 x 3분 동안 담그고 슬라이스를밀봉합니다 25.</li>
</ol>9. 효소 결합 면역 흡착 분석법(ELISA)
<ol><li>제조업체의 지침에 따라 각 키트를 사용하여 혈장 내 IL-17 및 TNF-α 농도를 측정합니다.</li>
</ol>10. 통계 분석
<ol><li>데이터를 평균± 평균의 표준 오차(SEM)로 표현합니다.</li>

강직성 척추염(AS)은 주로 축 골격에 영향을 미치는 점진적으로 악화되고 무력화되는 형태의 관절염입니다. 최신 역학 조사에 따르면, 전 세계 인구의 ~0.01%-1.8%가이 질환을 앓고 있으며1, 그 중 대부분은 젊은 남성입니다(남녀 비율은 ~2-3:1)2. 중국 인구의 AS 발생률은 0.2%-0.4%입니다. 이 질병은 주로 척추와 천장관절을 침범한다. 말기에는 척추와 천장관절의 유합이 일어나 척추 경직과 후만증, 보행의 어려움 등이 발생할 수 있으며, 이는 업무의 질과 일상생활에 심각한 영향을 미치고 환자와 가족, 사회에 큰 부담을 준다1. 현재 AS에 대한 확실한 해결책은 없으며, 제공되는 약리학적 및 비약물적 치료법은 주로 통증 완화, 질병 진행 지연 및 삶의 질 향상에 중점을 둡니다. 최근 몇 년 동안 약물 요법 옵션이 매우 제한적이었기 때문에3 AS의 대체 요법이 된 비약물 요법에 대한 관심이 높아지고 있습니다. 중국에서는 환자들이 종종 중국 전통 의학(TCM)으로 구성된 외부 치료를 선호하는데, 이는 부작용이 적고 치료 중 편의성이 매우 높습니다.
뜸은 한의학에서 사용되는 고대 치료 기법입니다. Du-moxibustion은 Du meridian의 척추 분절에 "거품이 있는 약용 뜸"으로 구성된 외부 치료 기술입니다. 주로 AS 치료에 사용되며 안전성과 효과를 보여주었습니다. 일반 뜸에 비해 두뜸은 넓은 뜸 면적, 큰 뜸 원뿔, 강한 화력 및 고온4의 특성을 가지고 있습니다. 체계적 문헌고찰 및 메타 분석에 따르면 뜸은 AS5에 대한 효과적인 보완 치료법이다. 최근 연구에 따르면 AS 환자의 임상 증상과 일부 염증 요인이 뜸에 의해 개선되는 것으로 확인되었다6. 연구에 따르면 듀뜸은 뇌에서 β-엔돌핀의 함량을 증가시키고 중추 진통 효과를 발휘하며7 HLA-B27 유전자의 발현을 하향 조절하고8 AS의 재발률을 지연시킬 수 있다. Du-moxibustion은 또한 염증 지수 ESR(ESR)과 C-반응성 단백질(CRP), I형 콜라겐의 C-말단 펩타이드(CTX-I) 및 Dickkopf1 단백질(DKK1)9,10의 수준을 감소시켜 AS의 진행을 개선하는 효과가 있습니다.
