Combined In Vivo Electroporation and Short&#45;Term Reinnervation of the Cranial Levator Auris Longus Skeletal Muscle

tdTomato 단백질20의 높은 형광 양자 수율은 전기천공법 효율에 적합한 추적자입니다. 따라서, in vivo electroporation에 의한 LAL 근육에서의 tdTomato 단백질의 발현을 평가하였다(그림 1). LAL 근육은 전기천공법 21일 후에 절개하였다. 전체 마운트 준비는 두 개의 신경 분포 영역이 있는 LAL 근육의 얇은 꼬리 밴드(cLAL, 청록색 다각형)와 5개의 신경 분포 영역 8,10이 있는 두꺼운 로스트랄 밴드(rLAL, 노란색 다각형)를 보여줍니다(그림 1B). 안면 신경의 후이변(posterior auricular branch)이 으스러지면 LAL 근육이 변성된다(그림 2A). 대조군 마우스와 신경 손상 후 5일, 10일 및 60일 후의 마우스로부터의 LAL 근육을 이전에 설명된 방법16,21을 사용하여 절개, 고정 및 염색하였다. 간단히 말해서, SV2 항체는 시냅스 소포를 표지하고 신경 필라멘트에 대한 2H3 항체와 함께 사용되어 전체 시냅스 전 도메인 6,16,22,23,24를 나타냅니다.
