Measuring Carbon Content in Airway Macrophages Exposed to Carbon-Containing Particulate Matters

고정액으로 보존 및 처리된 가래는 형태학적 검사 중에 광학 현미경 하에서 온전한 대식세포 형태를 보여주었습니다. 대식세포는 투명하고 둥글거나 신장 모양이며 쉽게 염색할 수 있는 세포핵을 나타냈습니다. 염색 후 세포핵은 청자색으로 보였고 세포질은 연한 분홍색 또는 연한 파란색이었습니다. 현미경 필드는 최소한의 불순물을 보여 주어 세포 식별이 용이했습니다. 세포 내에서 블랙 카본 입자가 작은 응집체에서 뚜렷하게 보였기 때문에 탄소 입자의 정량적 분석이 가능했습니다.
가래 대식세포의 대표적인 이미지가 그림 1에 제시되어 있으며, 서로 다른 세포에 있는 다양한 수의 탄소 입자를 보여줍니다. 예를 들어, 그림 1A는 탄소 입자가 없음을 보여주는 반면 그림 1D는 가장 높은 농도를 나타냅니다. CCAM의 값은 대식세포(A-D)에 대해 각각 0.00%, 2.24%, 6.64% 및 34.61%로 측정되었습니다. CCAM 매개변수 결정 프로세스 동안, 대식세포 크기의 변화 및 탄소 입자와 세포핵의 중복으로 인해 발생하는 편향을 완화하기 위해 각 대식세포의 탄소 함량에 대한 정량적 평가를 수행했습니다. CCAM 계산 및 분석은 그림 2와 같이 연구 대상자 당 50개의 대식세포에 대해 수행되었습니다.
