Utilizing Thermal Shift Assay to Probe Substrate Binding to Selenoprotein O

진핵생물 SelO는 N-말단 미토콘드리아 표적 서열, 키나아제와 유사한 도메인 및 단백질23의 C-말단에서 고도로 보존된 셀레노시스테인으로 구성됩니다. 이 미토콘드리아 상주 효소는 박테리아에서 인간에게 보존되는 슈도키나아제 도메인을 암호화한다23. 슈도모나스 시린가(Pseudomonas syringae)의 SelO 상동체(homolog)에 대한 구조 분석은 표준 키나아제(canonical kinases)와 비교하여 역방향(inverted orientation)으로 ATP의 결합을 촉진하는 활성 부위의 아미노산 변화를 밝혀냈다25. 따라서 SelO는 항산화 신호 전달에 관여하는 여러 단백질의 인산화가 아닌 AMPylation을 촉매하여 산화 손상 및 세포 사멸로부터 세포를 보호합니다25. AMPylation 외에도 일부 원핵 및 진핵 AMPylases가 UMPylation27,28을 촉매하는 것으로 나타났습니다.
TSA에서 정제된 재조합 E. coli SelO를 사용하여 2가 양이온이 있는 상태에서 ATP 및 UTP 뉴클레오티드와 SelO의 결합을 조사했습니다. 표준 키나아제와 유사하게, SelO는 아스파르테이트가 금속 이온과 결합하여 활성 부위의 뉴클레오티드를 조정하는 DFG 모티프를 가지고 있습니다. 먼저, TSA에서 강력한 형광 측정을 위해 단백질 및 염료의 농도를 최적화하여 5x SYPRO Orange에서 5μM - 10μM E. coli SelO가 최적임을 확인했습니다(그림 2). 또한, 단백질이 없는 경우와 염료가 없는 대조군은 온도 상승에 반응하지 않는 낮은 형광 신호를 가지고 있음을 보여주었습니다.
