Infection of In Vivo and In Vitro Pines with the Pinewood Nematode Bursaphelenchus xylophilus and Isolation of Induced Volatiles

PWN은 최적의 조건에서 빠르게 번식하며 생성 시간은 4일 정도로 짧을 수 있으며 각 암컷은 일생 동안 약 80개의 알을 낳습니다28. 상술한 방법론을 사용하여, 진균 성장에 따라 많은 양의 PWN을 얻을 수 있다. 8일의 성장 기간 내에 PWN은 개체 수가 100배 증가할 수 있습니다(그림 1). PWN 양의 일관성을 높이려면 알려지지 않은 박테리아 또는 곰팡이에 의한 오염이 PWN 개체군에 부정적인 영향을 미치므로 멸균된 PWN을 사용하십시오.
소나무 묘목이 PWN에 성공적으로 감염되면 감염 후 약 20일 후에 PWD 증상이 나타납니다(그림 2). 그러나 P. pinaster 는 PWD에 대한 감수성에 대한 유전적 변이가 높은 것으로 알려져 있으며, 증상 발현 시기에 대한 변동은 매우 흔하다3.
시험관 내 소나무 새싹은 이러한 유전적 다양성을 가지고 있지 않으며 환경 및 영양 조건은 사용자의 필요에 따라 제어되고 설정됩니다. 시험관 내 해양 소나무 싹 배양은 SH 신장 배지에서 잘 성장하여 주당 약 0.9mm의 성장률을 보인다6. 이상적으로는 멸균 PWN을 최소 5cm(바닥에서 가장 높은 바늘까지)의 새싹에 추가할 수 있습니다. 체외 소나무 싹이 더 작고 PWN 감염 면적이 줄어들기 때문에 선충 개체수 증가는 발현력이 떨어지므로 두 배로 늘어나는 시간은6일에서 7일 사이가 될 수 있다6. 그럼에도 불구하고, 클로로틱 바늘은 공동 배양 후 한 달 이내에 관찰될 수 있습니다(그림 3). 온실에서 재배한 소나무와 비교했을 때, 체외 소나무 싹 배양은 PWD 증상의 동기화, 온도 및 광주기 조절, 영양 상태 조절 및 오염 방지를 가능하게 합니다.
2 년생 P. pinaster 묘목은 약 0.2 % ± 0.1 % (v / fresh weight)의 에센셜 오일 수율과 α-pinene, β-pinene 또는 δ-3-carene이 풍부한 휘발성 물질 프로필을 가지며 일정한 화학적 변동성을 가지고 있습니다. 현재 두 가지 주요 화학유형이 확인되었는데, 하나는 α-pinene과 β-pinene이 우세한 것이고 다른 하나는 α-pinene과 β-pinene이 δ-3-carene5와 우세를 공유하는 것입니다. 해안소나무 in vitro 새싹은 원하는 화학형에 따라 배양할 수 있으며, 휘발성 물질은 GC-MS를 통해 완전히 프로파일링됩니다. 증류 추출로 얻은 휘발성 물질의 휘발성 프로필은 에센셜 오일의 휘발성 프로필과 다를 수 있지만 P. pinaster에서는 유사합니다(그림 5A). 휘발성 추출 기법은 소나무 휘발성 대사 산물 프로필의 유도 변화를 평가하는 데 적합합니다. 휘발성 포획 기술은 소나무에서 방출되는 휘발성 물질에 대한 정보를 제공합니다. SPME 기술의 화학적 프로필은 다를 수 있지만 차이점은 일반적으로 식물 재료의 양과 포획된 휘발성 물질에 따라 다릅니다(그림 5B, C).
