Setting a Successful Sorting for Extracellular Vesicle Isolation

FSC 폴리스티렌 비드는 기기 설정 및 분류 조건을 검증하기 위해 분류되었습니다. FSC 폴리스티렌 비드는 100nm, 300nm, 500nm 및 900nm 범위의 형광 비드의 혼합물이며 FITC 채널에서 볼 수 있습니다. 그림 7A 는 정렬 전에 4개의 비드 모집단이 있는 SSC 로그 척도 대 FITC 로그 척도 점도를 보여줍니다. 100 nm, 300 nm 및 500 nm의 형광 집단을 게이트하고 분류했습니다. 분류된 beads는 초기 개념 증명으로 순도 및 농축을 위해 분석되었습니다(그림 7B). 모든 개체군은 분류 후 순수했습니다. 참조 배경 소음은 정렬 전후에 점도표에서 항상 볼 수 있습니다.
2023년 MISEV2018에 나열된 요구 사항을 충족하고 소포의 특성과 순도를 입증하기 위해 EV에 존재하거나 없는 마커를 분석하기 위한 실험이 수행되었습니다. EV는 일부 특정 표면 마커15: CD9, CD63, CD81 및 CD44에 대해 양성입니다. CD9, CD63, CD81은 테트라스파닌 계열의 막 단백질에 속하며 엑소좀을 식별하는 가장 일반적인 마커입니다16. CD44는 MSC 및 ASC17에 의해 많이 표현되는 마커입니다.
EV는 또한 세포 내 유세포 분석 실험에서 입증된 바와 같이 세포질 단백질 Caveolin에 대해 양성입니다. 또한 웨스턴 블롯은 CD9, CD81, TSG101, Flotillin에 대한 양성 반응과 Calnexin에 대한 양성 반응을 확인시켜 줍니다(그림 2).
다음으로, CFSE 라벨링된 ASC-EV는 프로토콜에 보고된 대로 분류되었습니다. 이 분류 절차를 4회 반복하여 시료 준비, 염색 및 분류에 대해 정확히 동일한 조건을 유지했습니다(표 1). 이러한 방식으로 프로토콜의 반복성이 입증되었습니다.
CFSE로 분류된 ASC-EV 모집단을 분석하고 분류에 의해 특성이 변경되지 않았음을 입증하기 위해 일련의 실험이 수행되었습니다. 특히, CD9, CD63, CD81 및 CD44의 발현은 분류 후에도 변하지 않습니다(그림 6A).
또 다른 중요한 점은 EV의 치수와 압력으로 인한 응력으로 인해 분류 절차 후에 어떻게 변경될 수 있는지에 관한 것입니다. 시료 양이 적어 분류 후 분석은 수행하기 어려웠지만, 나노입자 추적 분석은 두 ASC-EV 샘플(분류 전과 후)의 분포가 동일하다는 것을 보여줍니다(그림 6B). 마지막으로, TEM을 통한 EV의 분석은 분류 후에도 형태가 크게 변경되지 않은 것으로 보인다는 것을 보여줍니다(그림 6C).
주목할 점은 MIFlowCyt-EV 프레임워크 가이드라인18에서 제안한 바와 같이, 0.1% Triton X-100 시료 처리 시 CFSE 양성 EV의 양 감소가 관찰되어 소포 기원을 뒷받침한다는 것입니다(그림 8).
