4. 시험관 내 유비퀴틴 사슬 절단 분석
참고: 이전 보고서 31에 기술된 DUB 특이성을 기반으로 유비퀴틴 사슬 길이 및 연결 유형을 선택하십시오31 또는 경험적으로 결정된 것입니다. 필요한 경우, 이 프로토콜은 정의된 길이 및 연결 유형의 상업적으로 이용 가능한 유비퀴틴 사슬 패널에서 관심 있는 야생형 DUB의 활성을 테스트하는 데 사용할 수 있습니다. 375-750 ng의 디-유비퀴틴 사슬 양과 1-2 μM의 DUB 농도를 분석의 시작점으로 사용할 수 있습니다27.

USP27X 촉매 활성에 대한 XLID105 관련 변이체의 영향을 확인하기 위해 GST 태그가 부착된 야생형 USP27X 및 XLID105 관련 변이체 F313V, Y381H 및 S404N USP27X 단백질을 박테리아에서 정제했습니다. 이러한 변형체는 USP27X의 USP 촉매 도메인 내에 있습니다(그림 2A).
