Translation Efficiency Test Using Polysome Profiles Under Heat Stress

애기장대(Arabidopsis)의 야생형인 Col-0은 16시간:8시간의 광주기 미만의 MS 배지에서 성장했습니다. 제어를 위해 열 스트레스 처리 없이 5일 된 묘목을 사용했습니다. 열 스트레스 그룹은 예열된 수조에서 40°C에서 1시간 동안 열처리를 거친 반면, 회수 그룹은 열처리 직후 22°C에서 2시간 동안 배치했습니다. 다양한 열처리 조건과 회수 조건을 사용함으로써 후속 단계를 활용하여 병진 효율을 측정할 수 있습니다.

다양한 처리에서 5일된 Col-0 묘목에 대한 폴리솜 프로파일링 플롯이 그림 2에 나와 있습니다. 대조군의 결과는 비폴리솜 분획과 폴리솜 분획 사이의 명확한 분리를 보여줍니다(그림 2A). 그러나 40°C에서 1시간의 열 응력 후 폴리솜 분획의 신호 강도가 크게 감소했습니다(그림 2B). 열처리된 묘목을 22°C에서 2시간 동안 회복시키면 폴리솜 분획의 신호 강도가 대조군과 유사한 수준으로 회복되었습니다(그림 2C). 그 결과 1시간의 열스트레스 처리가 애기장대(Arabidopsis) 묘목의 번역 효율을 크게 감소시킨다는 것을 보여주었습니다. 그러나 식물은 열 스트레스를 받지 않는 묘목과 비슷한 수준으로 번역 효율을 복원하기 위해 2시간의 회복이 필요했습니다.
다양한 처리 조건에서 Col-0 묘목의 번역 효율을 비교하는 추가 증거를 수집하기 위해 비폴리솜 및 폴리솜 RNA 분획을 분리하고 추출했습니다. 비폴리솜 및 폴리솜 RNA 분획의 수집은 폴리솜 프로파일 측정 결과에 따라 안내되었습니다. 비폴리솜 및 폴리솜 RNA 추출의 경우 페놀/구아니디늄 티오시아네이트 시약을 사용하여 분리된 비폴리솜 및 폴리솜 RNA 샘플을 추출합니다. 우리가 사용한 RNA 추출 방법을 사용하면 다음과 같은 분석을 위한 안정적이고 고품질의 RNA를 얻을 수 있습니다(표 1). 그 후, GeneChip Eukaryotic Poly-A RNA Control Kit로 추출된 RNA를 정규화하여 다양한 추출 손실로 인한 오류를 완화합니다. 다음으로, 키트에 추가된 B. subtilis 유전자에 포함된 DAP 유전자의 발현을 정량화하기 위해 Real-Time PCR을 사용하였다. 비폴리솜 및 폴리솜 RNA 그룹 간의 DAP 유전자 발현 수준의 비교 분석을 수행하여 비례 손실 조건에서 RNA 함량을 추정했습니다(그림 3A). 우리는 그들 사이의 번역 효율성을 명확히하기 위해 총 RNA에 대한 정규화 된 폴리솜 RNA의 비율을 계산했습니다 (그림 3B). 마지막으로, 폴리솜의 RNA 비율은 정규화된 RNA 함량을 사용하여 계산되었습니다(그림 3C).
