Using the Droplet Transfer Method to Reliably Prepare Giant Unilamellar Vesicles

최적의 GUV 준비는 투명한 유상과 뚜렷한 펠릿의 형성을 특징으로 합니다(그림 4A). 물방울이 GUV로 변환되지 않는 경우 일반적으로 붕괴되어 OS/LS 인터페이스에 인지질 파편이 남습니다(그림 4C). 이 효과는 물/기름 계면에서 인지질의 불충분한 평형으로 인해 유상에서 인지질 및 기타 분자의 응집으로 인해 발생할 가능성이 큽니다. 그럼에도 불구하고 눈에 보이는 펠릿이 없거나 인터페이스에 파편이 있음에도 불구하고 GUV는 여전히 형성되어 프로세스를 계속할 수 있습니다. 세척 단계가 모든 인지질 잔해물을 제거하는 데 완전히 효과적인 것은 아니지만 오일 방울을 제거하는 데 크게 도움이 되어 전체 샘플 품질을 향상시킵니다( 그림 6A 및 그림 6B 비교).
고습도 조건에서 준비된 오래된 LS 또는 LS를 사용하면 GUV 분산액이 감소하고 수율이 낮아지고 유상의 인지질 및 기타 분자에 의해 형성될 가능성이 가장 높은 무질서한 응집체의 함량이 높아집니다(그림 6C). 고습도 조건에서 준비된 LS의 경우 GUV 생산(GUV 포함 샘플이 거의 없는 샘플)에서 0.8 이상의 변동 계수를 관찰한 반면, 수정된 장갑 챔버에서 준비된 LS의 경우 변동 계수는 0.5 미만이었습니다.
다양한 원심분리 조건은 GUV 수율과 품질에 영향을 미칠 수 있습니다. 정성 분석에서 3,300 × g에서 20분이 자당 로딩 GUV(샘플 E)에 대한 양호한 조건으로 확인되었습니다. 속도를 낮추면(500×g에서 45분) 수율이 감소한 반면, 증가(16,600×g에서 5분)하면 무질서한 인지질 응집체가 더 많이 형성되었습니다. 약한 원심력은 많은 물방울이 OS/LS 계면을 통과하는 것을 방지하여 수율을 감소시킬 수 있는 반면, 강한 힘은 계면 교차 중에 외부 전단지의 인지질 조직화에 충분한 시간을 제공하지 못할 수 있습니다(그림 7). 미세 입자의 캡슐화는 미세 입자의 질량 밀도가 IS의 질량 밀도와 대략 일치하는 경우 원심 분리 매개변수를 조정하여 최적화할 수도 있습니다. 이를 위해 원심분리 가속도를 낮추고(차례로 원심분리 시간을 늘리면) 고체 입자에 의해 가해지는 힘으로 인한 인지질 막의 파손을 방지함으로써 입자의 보다 균일한 캡슐화가 가능합니다(최적의 원심분리 매개변수에 대한 조사가 아직 진행 중인 동안 우수한 캡슐화의 예는 그림 6D에 나와 있습니다17).
인지질의 확산은 유상의 영향을 받을 수 있기 때문에 간단한 정성 테스트를 실행하여 에멀젼의 안정성을 평가할 수 있습니다(그림 5). 시각화를 개선하기 위해 IS에 용해된 착색 염료(Brilliant Blue FCF)를 사용하여 물/기름 비율이 다른 에멀젼을 준비하고 시간이 지남에 따라 관찰할 수 있습니다. 안정성 손실은 물방울의 응집과 그에 따른 유상으로부터의 분리로 시각화할 수 있습니다. 큰 물방울(육안으로 볼 수 있음)은 GUV로 전환될 가능성이 적기 때문에 상 분리가 가장 적은 에멀젼 조건을 사용하는 것이 좋습니다.
프로토콜의 변화의 효과는 프로토콜 섹션 4에 설명된 대로 크기 분포 분석을 통해 검증할 수 있습니다: GUV의 직경은 위상차 현미경 이미지에서 ImageJ를 사용하여 측정할 수 있으며 크기 분포는 GitHub16에서 사용할 수 있는 스크립트로 분석할 수 있습니다. 예를 들어, 이 프로토콜에 따라 실리콘 오일 AR20보다 미네랄 오일을 선택할 때 더 큰 직경의 GUV가 증가하는 것을 관찰할 수 있었습니다(그림 8).
