The Nucleus

- [Verteller] De kern, een opvallend kenmerk van eukaryotische cellen, is een membraangebonden organel dat genetische informatie herbergt, het DNA, die anders is georganiseerd, afhankelijk van op de levenscyclusfase van de cel. Bijvoorbeeld, DNA-eiwitcomplexen, bekend als chromatine, zijn gesuspendeerd in nucleoplasma, een gelachtige substantie, en zijn bijzonder geconcentreerd in een gebied die de nucleolus wordt genoemd. Wanneer de cel zich niet deelt, zijn de chromatinevezels losjes gerangschikt en lijken ze op verwarde strengen, waardoor tijdens de transcriptie gemakkelijker toegang tot het DNA mogelijk is.

Wanneer de cel zich daarentegen wel gaat delen, worden chromatinevezels tot chromosomen samengeperst. De nucleolus is ook waar de subeenheden van ribosomen, structuren die nodig zijn voor vertaling, worden gemaakt. Rondom de kern bevindt zich een dubbelmembraan nucleair omhulsel, dat met eiwit beklede poriën bevat, om de stroom van ionen, moleculen, ribosomale delen en boodschapper-RNA tussen het binnenste nucleoplasma en het cytoplasma te regelen.

Terwijl de meeste cellen in het lichaam één kern hebben, sommige zoals rode bloedcellen zijn geanucleeerd, missen ze een kern. Terwijl anderen, zoals skeletspiercellen, meerkernig zijn, met meer dan één kern, vanwege structurele en functionele behoeften.

De kern is een membraangebonden organel dat fungeert als het controlecentrum van een eukaryotische cel. Het bevat chromosomaal DNA, dat de genexpressie reguleert en de eiwitproductie in de cel nauwkeurig controleert. Daarentegen voert het DNA in de mitochondriën en chloroplasten alleen functies uit die specifiek zijn voor die organellen.

<h4>Regeling van DNA binnen de kern</h4>

De regulatie van genexpressie in de kern hangt af van verschillende factoren, waaronder de DNA-structuur. Het DNA in de kern is gewikkeld rond eiwitten die histonen worden genoemd, wat resulteert in een DNA-eiwitcomplex dat chromatine heet. Wanneer cellen zich niet delen (tijdens de interfase), is het chromatine diffuus georganiseerd. Dit maakt gemakkelijke toegang tot het DNA mogelijk tijdens transcriptie. Wanneer de eukaryote cel zich voorbereidt op deling, condenseert het chromatine in strak georganiseerde, lineaire chromosomen, die eenvoudig kunnen worden gescheiden in dochtercellen.

<h4>Nucleoli</h4>

De nucleolus is een membraanloos organel dat zich vormt rond de chromosomale loci van het ribosomale RNA (rRNA) in de kern. Daarom zijn nucleoli de primaire locatie voor rRNA-synthese en de assemblage van ribosomale subeenheden. Deze subeenheden worden later uit de kern geëxporteerd voor assemblage tot volledig functionele ribosomen.

<h4>Aanvullende subnucleaire structuren</h4>

De kern bevat ook verschillende andere nucleaire lichamen, zoals Cajal-lichamen, spikkels en paraspeckles. Deze nucleaire lichamen helpen bij de ruimtelijke compartimentering van de nucleaire omgeving en creëren gespecialiseerde locaties voor specifieke biologische reacties. Bovendien bevordert hun membraanloze structuur een snellere uitwisseling van inhoud met het nucleoplasma.

The nucleus is the control center of a eukaryotic cell. It is a membrane-bound organelle that contains chromosomal DNA. That DNA helps control gene expression and regulate protein production in the cell.

Not all cellular DNA has the same role. The DNA in mitochondria and chloroplasts supports functions that are specific to those organelles. Inside the nucleus, DNA structure also affects how genes are regulated.

Nuclear DNA is wrapped around proteins called histones. This forms chromatin, a DNA-protein complex. During interphase, when the cell is not dividing, chromatin stays loosely arranged. This open form makes the DNA easier to access during transcription.

When a eukaryotic cell prepares to divide, chromatin changes shape. It condenses into distinct, linear chromosomes. These chromosomes can then be separated into daughter cells.

The nucleus also contains the nucleolus. This is a membrane-less organelle that forms around the chromosomal loci for ribosomal RNA, or rRNA. The nucleolus is the site where rRNA is made and ribosomal subunits are assembled. These subunits leave the nucleus and later combine to form functional ribosomes.

Other nuclear bodies are found in the nucleus as well. These include Cajal bodies, speckles, and paraspeckles. They help organize the nuclear space and create separate sites for specific biological reactions. Their membrane-less structure also allows contents to move more easily with the nucleoplasm.
