Mutations

- Puntmutaties, waarbij &#xE9;&#xE9;n enkele nucleotide veranderd wordt, kunnen een prote&#xEF;ne produceren dat compleet normaal is, of totaal niet-functioneel, afhankelijk van het type.
 Deze omvatten stille, missense, nonsense en frameshift mutaties.
 Stille mutaties, zoals de naam al zegt, hebben geen effect op de aminozuursequentie van een prote&#xEF;ne.
Bijvoorbeeld, als het codon CCA veranderd wordt naar CCG, zal het nog steeds coderen voor het aminozuur proline en de prote&#xEF;ne zal normaal functioneren.
 Missense mutaties anderzijds resulteren in de substitutie van een aminozuur voor een ander, zoals arginine in plaats van proline, waardoor de prote&#xEF;ne slecht kan gaan functioneren.

Gevolgen van puntmutaties op moleculair niveau
Mutaties die &#xE9;&#xE9;n nucleotide aantasten, worden puntmutaties genoemd. Wanneer puntmutaties binnen genen ontstaan, kunnen de gevolgen in ernst vari&#xEB;ren, afhankelijk van de manier waarop ze de gecodeerde aminozuursequentie veranderen. Een stille mutatie verandert de aminozuursequentie niet en is vaak neutraal. Een missensemutatie verandert &#xE9;&#xE9;n aminozuur en de gevolgen kunnen ernstig zijn als de verandering de functie van het eiwit verandert. Een nonsensemutatie introduceert een stopcodon, waardoor het eiwit wordt verkort en waarschijnlijk niet-functioneel wordt. Chromosomale veranderingen zijn grootschalige mutaties

Structurele veranderingen kunnen bestaan uit deleties, duplicaties, inversies en translocaties van grote DNA-segmenten binnen &#xE9;&#xE9;n chromosoom. Numerieke afwijkingen, zoals aneuplo&#xEF;die, omvatten de winst of het verlies van volledige chromosomen. Chromosomale mutaties zijn doorgaans veel ernstiger dan puntmutaties, omdat ze veel genen en regulatoire elementen be&#xEF;nvloeden. Chromosomale veranderingen kunnen worden opgespoord door karyotypering van de aangetaste cel.
Alleen kiemlijnmutaties worden overge&#xEB;rfd
Mutaties kunnen in elke cel ontstaan, maar alleen kiemlijnmutaties&#x2014;mutaties die aanwezig zijn in eicellen en zaadcellen&#x2014;kunnen aan nakomelingen worden doorgegeven. Erfelijke ziekten zijn een subtype van genetische aandoeningen die worden veroorzaakt door schadelijke kiemlijnmutaties. Ze kunnen autosomaal zijn, waarbij de chromosomen 1 tot en met 22 betrokken zijn, of geslachtsgebonden, waarbij het X- of Y-chromosoom betrokken is. Een voorbeeld van een erfelijke ziekte is cystische fibrose (CF), die voornamelijk de longen aantast. Deze ziekte wordt veroorzaakt door een deletie in het CFTR-gen die &#xE9;&#xE9;n aminozuur verwijdert uit het CFTR-eiwit. CF is een autosomaal recessieve ziekte, wat betekent dat iemand met &#xE9;&#xE9;n gemuteerde kopie van het gen en &#xE9;&#xE9;n normale kopie de ziekte niet zal ontwikkelen. Hiertoe behoren omgevingsstraling of chemische stoffen die DNA beschadigen. Een goed gekarakteriseerd omgevingsmutageen is ultraviolette (UV) straling. UV-straling induceert de vorming van covalente bindingen tussen aangrenzende thyminebasen op dezelfde DNA-streng, waardoor thyminedimeren ontstaan die de DNA-structuur vervormen. De zon is een natuurlijke bron van UV-straling. De schadelijkste UV-C-stralen worden door de atmosfeer geabsorbeerd, terwijl UV-A- en UV-B-stralen het aardoppervlak bereiken. Kunstmatige bronnen van UV-blootstelling zijn onder meer zonnebanken, die voornamelijk UV-A-stralen en kleinere hoeveelheden UV-B uitzenden.
Gelukkig beschikken cellen over mechanismen om beschadigd DNA te herstellen, maar soms wordt de schade niet v&#xF3;&#xF3;r de celdeling hersteld, met name in zich snel delende cellen, zoals huidcellen, en kunnen mutaties zich ophopen. Wanneer deze mutaties genen aantasten die celgroei en celdeling reguleren, kunnen ze bijdragen aan de ontwikkeling van kanker.

Mutations are changes in the DNA sequence. They can happen by chance during DNA copying, or they can be caused by environmental factors such as radiation and chemicals. Scientists also describe mutations by how much DNA they affect, how they change a protein, and whether they happen in body cells or germline cells.

Some mutations affect only one nucleotide, which is a point mutation. When a point mutation falls inside a gene, the outcome depends on how the amino acid sequence changes. A silent mutation leaves the amino acid the same and usually has no effect. A missense mutation changes one amino acid, and the result can be serious if the protein no longer works well.

Other point mutations can stop protein production early. A nonsense mutation creates a stop codon, which truncates the protein and often makes it nonfunctional. A frameshift mutation happens when nucleotides are inserted or deleted in protein-coding DNA. This shifts the reading frame and changes all downstream codons.

Some mutations are large-scale chromosomal alterations. These changes affect the physical structure of a chromosome and can include deletion, duplication, inversion, or the addition of DNA from another chromosome. Because they can involve many genes and regulatory elements, they are usually more serious than point mutations. Cells with these changes can be identified by karyotyping.

Only germline mutations are inherited. These mutations are present in egg and sperm cells and can pass to offspring, while somatic mutations stay outside the germline. Hereditary diseases are caused by harmful germline mutations. They can be autosomal, on chromosomes 1 through 22, or sex-linked, on the X or Y chromosome.

Cystic fibrosis is one example of a hereditary disease. It affects the lungs and is caused by a deletion in the CFTR gene that removes a single amino acid from the CFTR protein. It is autosomal recessive, so one normal copy can prevent disease. Huntington's disease is another example, and it is autosomal dominant, which means one mutated copy is enough for the disorder to develop.

Mutations can also come from mutagens, which are external agents that damage DNA. Ultraviolet, or UV, radiation is a well-known mutagen. It has more energy than visible light and can create thymine dimers when bases on the same DNA strand bond with each other. The sun is a natural source of UV radiation, and tanning beds are artificial sources that mainly emit UV-A with some UV-B.

Cells can repair DNA damage, but repair is not always complete before cell division. This matters most in rapidly dividing cells such as skin cells. If the damage affects genes that help control cell growth and division, it can contribute to cancer.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
