The DNA Helix

- [Narrator] Deoxyribonucleïnezuur of DNA, bestaat uit twee polynucleotide ketens. Elk bestaat uit vier soorten nucleotidesubeenheden, die bestaan uit een enkele fosfaatgroep, en een stikstof bevattende basis, Adenine A, Cytosine C, Guanine G of Thymine T. Deze opstelling is gehecht aan een suiker deoxyribose, en vormt de kern van suikerfosfaat van de DNA-opbouw.

Aan één keten, bij de drie-prime end, is de suiker georiënteerd met de hydroxylgroep blootgesteld bij het derde koolstofatoom die klaar is om covalent te koppelen aan een andere suikermolecuul in de keten bij het vijfde koolstofatoom via fosfodiesterbindingen. In de tegenovergestelde anti-parallelle streng, is elke basis uniek gepaard, C met G, en A met T, dankzij verschillen in het aantal waterstofbruggen die tussen hen vormen. Deze complementaire zijde eindigt met een fosfaatgroep aan het eerste vijfpunt.

Een dergelijke chemische polariteit zorgt voor een efficiënte verpakking, met de twee strengen om elkaar heen om een 3D dubbele helix te vormen met tien basissparen iedere volledige beweging.

<h4>Overzicht</h4>

Deoxyribonucleïnezuur (DNA) is het genetische materiaal dat verantwoordelijk is voor het doorgeven van eigenschappen van generatie op generatie in alle organismen en de meeste virussen. DNA bestaat uit twee strengen nucleotiden die om elkaar heen wikkelen en zo een dubbele helix vormen. De ontdekking van de structuur van DNA gebeurde stapsgewijs gedurende bijna een eeuw en vertegenwoordigde een van de beroemdste en meest boeiende verhalen in de geschiedenis van de wetenschap.

<h4>DNA-structuur in detail</h4>

Elke DNA-streng bestaat uit subeenheden die nucleotiden worden genoemd en die de suiker deoxyribose, een fosfaatgroep en een van de vier stikstofbevattende basen bevatten: adenine (A), guanine (G), cytosine (C) en thymine (T). Adenine en guanine behoren tot een grotere klasse chemicaliën, purines genaamd, die allemaal structuren met twee ringen bevatten. Cytosine en thymine behoren tot een groep enkelringige structuren die pyrimidines worden genoemd.

Aangrenzende nucleotiden in dezelfde streng zijn covalent verbonden door fosfodiësterbindingen. De twee strengen nucleotiden worden bij elkaar gehouden door waterstofbruggen, waarbij de adenines in de ene streng paren met thyminen op dezelfde positie in de andere streng, en de cytosines in de ene streng paren met guanines op dezelfde positie in de andere streng.Deze waterstofbinding wordt mogelijk gemaakt door de antiparallelle opstelling van de twee DNA-strengen, waarbij de 5’ en 3’ uiteinden van de strengen in tegengestelde richtingen zijn georiënteerd. Zonder deze opstelling zouden de nucleotiden zich in de verkeerde positie bevinden om waterstofbruggen tussen strengen te vormen.

De twee strengen van het DNA-molecuul zijn strak gewikkeld in een veerachtige structuur die een dubbele helix wordt genoemd. De dubbele helix is echter niet perfect symmetrisch. In plaats daarvan zijn er regelmatig voorkomende groeven in de constructie. De grote groef ontstaat daar waar de suiker-fosfaatruggengraat relatief ver uit elkaar ligt, wat toegang geeft tot DNA-bindende eiwitten, zoals transcriptiefactoren. De kleine groef komt daarentegen voor waar de suiker-fosfaatruggengraat dicht bij elkaar ligt. Relatief weinig eiwitten binden via de kleine groef aan DNA.

<h4>De ontdekking van de DNA-structuur: een korte geschiedenis</h4>

Het verhaal van de ontdekking van de DNA-structuur begint in 1869 toen de Zwitserse wetenschapper Friedrich Miescher een stof ontdekte die hij 'nucleïne' noemde. Tijdens het extraheren van eiwitten uit witte bloedcellen ontdekte Miescher een onverwachte stof met een relatief hoog fosforgehalte. Hij wist niet wat het was, maar hij vermoedde dat het biologisch belangrijk zou kunnen zijn. Miescher had gelijk, maar het duurde tientallen jaren voordat de wetenschappelijke gemeenschap zijn inzichten ten volle kon waarderen.

De volgende kritische ontdekking werd gedaan door de Russische biochemicus Phoebus Levene. In 1919 stelde Levene voor dat nucleïne, toen bekend als nucleïnezuur, bestond uit ketens van moleculen die hij polynucleotiden noemde. Het voorstel van Levene kwam voort uit zijn onderzoek naar gist, waarin hij ontdekte dat individuele nucleotiden waren opgebouwd uit een fosfaatgroep, een suiker en een stikstofhoudende base. Hoewel het polynucleotidemodel van Levene in veel opzichten correct was, bleef onduidelijk hoe de basen in het DNA-molecuul waren gerangschikt.

De Oostenrijkse biochemicus Erwin Chargaff borduurde voort op het werk van Levene. Chargaff deed eind jaren veertig een belangrijke ontdekking: de hoeveelheid adenine in DNA is altijd ongeveer gelijk aan de hoeveelheid thymine, en de hoeveelheid guanine is altijd ongeveer gelijk aan de hoeveelheid cytosine. Dit patroon werd bekend als de regel van Chargaff en was een belangrijk bewijsstuk dat de uiteindelijke opheldering van de DNA-structuur mogelijk maakte.

Begin jaren vijftig waren de Amerikaanse bioloog James Watson en de Engelse natuurkundige Francis Crick in een race met hun belangrijkste rivaal, de Amerikaan Linus Pauling, om de driedimensionale structuur van DNA te ontdekken. Voortbouwend op het werk van Chargaff gebruikten ze kennis van natuurkunde, wiskunde en scheikunde om fysieke modellen van DNA te construeren, maar ze waren niet succesvol totdat ze een cruciaal stukje gegevens ontvingen: een röntgenfoto van DNA dat in nauwkeurig detail de dubbele helixstructuur aangaf. Deze foto was de ongepubliceerde gegevens van natuurkundige Rosalind Franklin en werd zonder medeweten van Franklin aan Watson en Crick gegeven. Watson en Crick publiceerden hun beschrijving van de DNA-structuur in 1953, en samen met Maurice Wilkins (een medewerker van Franklin) wonnen ze in 1962 de Nobelprijs voor de Fysiologie of Geneeskunde voor deze ontdekking. Helaas stierf Franklin in 1958 en kwam daarom niet in aanmerking voor een Nobelprijs.
