The Central Dogma

- [Verteller] Het centrale dogma van de moleculaire biologie stelt dat informatie die gecodeerd is in het DNA wordt overgebracht naar RNA, dat vervolgens de synthese van eiwitten stuurt op basis van deze instructies. Ten eerste, in het proces van transcriptie, wordt DNA gebruikt als een voorbeeld om boodschapper-RNA, mRNA, te synthetiseren, wat een kopie van de coderingsstreng is. Behalve dat de thymidines zijn vervangen door uracils.

Bij de volgende stap in het transleerproces in eukaryoten, reist mRNA naar een ribosoom. Hier binden codons, groepen van drie nucleotiden in het mRNA, zich aan complementaire sequenties op transfer-RNA, tRNA, moleculen, die elk verbonden zijn aan een bepaald aminozuur, afhankelijk van de specifieke codon. Bijvoorbeeld, de codon CCA bindt aan een tRNA die aan de proline is bevestigd, terwijl AGC bindt aan een tRNA die aan serine is bevestigd.

Op deze manier geeft de genetische code de volgorde aan waarin de aminozuren in het resulterende polypeptide worden gerangschikt. Polypeptiden worden dan vaak verder verwerkt tot functionele eiwitten.

<h4>Overzicht</h4>

Het centrale dogma van de biologie stelt dat informatie die in het DNA is gecodeerd, wordt overgebracht naar messenger RNA (mRNA), dat vervolgens de synthese van eiwitten aanstuurt. De reeks instructies waarmee de mRNA-nucleotidesequentie in aminozuren kan worden gedecodeerd, wordt de genetische code genoemd. Het universele karakter van deze genetische code heeft geleid tot vooruitgang in wetenschappelijk onderzoek, landbouw en geneeskunde.

<h4>RNA is de ontbrekende schakel tussen DNA en eiwitten</h4>

Aan het begin van de 20e eeuw ontdekten wetenschappers dat DNA alle informatie opslaat die nodig is voor cellulaire functies en dat eiwitten de meeste van deze functies vervullen. De mechanismen voor het omzetten van genetische informatie in functionele eiwitten bleven echter jarenlang onbekend. Aanvankelijk werd aangenomen dat een enkel gen direct wordt omgezet in het gecodeerde eiwit ervan. Twee cruciale ontdekkingen in eukaryote cellen betwistten deze theorie: ten eerste vindt de eiwitproductie niet plaats in de kern. Ten tweede is DNA niet aanwezig buiten de kern. Deze bevindingen leidden tot de zoektocht naar een intermediair molecuul dat DNA verbindt met de eiwitproductie. Dit intermediaire molecuul, dat zowel in de kern als in het cytoplasma wordt aangetroffen en geassocieerd is met de eiwitproductie, is RNA.

Tijdens transcriptie wordt RNA in de kern gesynthetiseerd, waarbij DNA als sjabloon wordt gebruikt. Het nieuw gesynthetiseerde RNA is qua sequentie vergelijkbaar met de DNA-streng, behalve dat thymidine in DNA wordt vervangen door uracil in RNA. In eukaryoten wordt dit primaire transcript verder verwerkt, waarbij de niet-coderende eiwitgebieden worden verwijderd, waardoor de 5’ eindigen en een 3’ poly-A-staart, om mRNA te creëren dat vervolgens naar het cytoplasma wordt geëxporteerd.

<h4>De regels voor het interpreteren van de mRNA-sequentie vormen de genetische code</h4>

Translatie vindt plaats bij ribosomen in het cytoplasma, waar informatie gecodeerd in het mRNA wordt vertaald in een aminozuurketen. Een set van drie nucleotiden codeert voor een aminozuur en deze tripletten worden codons genoemd. De reeks regels die schetsen welke codons een bepaald aminozuur specificeren, vormen de genetische code.

<h4>De genetische code is overbodig</h4>

Eiwitten worden opgebouwd uit 20 aminozuren in eukaryoten. Het combineren van vier nucleotiden in sets van drie levert 64 (43) mogelijke codons op. Dit betekent dat het mogelijk is dat een individueel aminozuur door meer dan één codon kan worden gecodeerd. Er wordt gezegd dat de genetische code overbodig of gedegenereerd is. Vaak, maar niet altijd, verschillen codons die dezelfde aminozuren specificeren alleen in het derde nucleotide van het triplet. De codons GUU, GUC, GUA en GUG vertegenwoordigen bijvoorbeeld allemaal het aminozuur valine. AUG is echter het enige codon dat het aminozuur methionine vertegenwoordigt. Het codon AUG is tevens het codon waar de eiwitsynthese start en wordt daarom het startcodon genoemd. Redundantie in het systeem minimaliseert de schadelijke effecten van mutaties. Een mutatie (d.w.z. verandering) op de derde positie van het codon hoeft niet noodzakelijkerwijs te resulteren in een verandering van het aminozuur.

<h4>De genetische code is universeel</h4>

Op enkele uitzonderingen na gebruiken de meeste prokaryote en eukaryote organismen dezelfde genetische code voor eiwitsynthese. Deze universaliteit van de genetische code heeft vooruitgang mogelijk gemaakt in wetenschappelijk onderzoek, landbouw en geneeskunde. Zo kan humane insuline nu op grote schaal in bacteriën worden geproduceerd. Dit gebeurt met behulp van recombinant-DNA-technologie. Recombinant DNA bestaat uit genetisch materiaal van verschillende soorten. Genen die voor menselijk insuline coderen, worden samengevoegd met bacterieel DNA en in een bacteriële cel ingebracht. De bacteriële cel voert transcriptie en translatie uit om de menselijke insuline te produceren die in het recombinante DNA wordt gecodeerd. De resulterende humane insuline wordt gebruikt om diabetes te behandelen.
