- [Instructeur] De meeste prokaryotische factoren benut tijdens replicatie hebben equivalenten die vergelijkbare rollen spelen in eurkaryotische DNA duplicatie.
 Dit proces initieert bij het begin van replicatie aan welke een herkenningscomplex bindt.
 Helicase is dan aangetrokken aan de locatie en scheidt de DNA strengen door een bubbel te genereren met twee vorken.
Primase komt ook en genereert RNA primers welke, terwijl helicase verder beweegt, de DNA polymerase verlengt met nieuw DNA.
 Net als in prokaryoten, de nieuw gevormde leidende streng groeit continu door de nieuwe replicatie vork te volgen.
 Daarentegen wordt de achterblijvende streng gevormd in kleine Okazaki fragmenten in de tegenovergestelde vork.
Door meerdere factoren, wordt het DNA model gebruikt om de leidende streng in een helft van deze structuur te genereren gebruikt om de achtergebleven streng in de ander te cre&#xEB;ren.
 Interessant is dat er verschillende replicatie herkomsten bestaan op een lineair eukaryotisch chromosoom, en replicatie eindigt wanneer hun geassocieerde gebieden samensmelten.
 Primers worden dan ge&#xEB;limineerd via enzymen zoals RNAse en uitgewisseld voor DNA.
Daarna bevestigt DNA ligase zich aan de segmenten.
 Alhoewel, wanneer de eind primer verdwijnt van de achterblijvende streng, blijft de ruimte leeg en is er een ongekopieerde strook van het DNA model aangrenzend.
 Om dit tegen te gaan, wordt een enzym genaamd telomerase aan de overhangende regio vastgehecht en verlengt het met een niet-coderende DNA sequentie.
Primase en DNA polymerase handelen deze verlengde regio en cre&#xEB;ren een telomeer kap die beschermt tegen het verlies van coderend DNA van de achterblijvende streng tijdens meerdere replicaties.
 Dus eindigt eurkaryotische DNA replicatie met twee DNA moleculen, elk met een ouderlijke en nieuw gesynthetiseerde streng, meerdere herkomsten van replicatie, en telomeren.

In eukaryotische cellen is de DNA-replicatie in hoge mate geconserveerd en strak gereguleerd. Meerdere lineaire chromosomen moeten v&#xF3;&#xF3;r de celdeling met hoge betrouwbaarheid worden gedupliceerd, dus er zijn veel eiwitten die een gespecialiseerde rol vervullen in het replicatieproces. Replicatie vindt plaats in drie fasen: initiatie, verlenging en be&#xEB;indiging, en eindigt met twee volledige sets chromosomen in de kern.

<h4>Veel eiwitten orkestreren de replicatie bij de oorsprong</h4>

Eukaryotische replicatie volgt veel van dezelfde principes als prokaryotische DNA-replicatie, maar omdat het genoom veel groter is en de chromosomen lineair zijn in plaats van circulair, vereist het proces meer eiwitten en kent het een paar belangrijke verschillen. Replicatie vindt gelijktijdig plaats op meerdere replicatieoorsprongen langs elk chromosoom. Initiatoreiwitten herkennen en binden zich aan de oorsprong, waarbij ze helicase rekruteren om de dubbele DNA-helix af te wikkelen. Op elk punt van oorsprong vormen zich twee replicatievorken. Primase voegt vervolgens korte RNA-primers toe aan de afzonderlijke DNA-strengen, die dienen als startpunt voor DNA-polymerase om te binden en de sequentie te kopi&#xEB;ren. DNA kan alleen worden gesynthetiseerd in de 5&#x2019; naar 3&#x2019; richting, dus de replicatie van beide strengen vanuit een enkele replicatievork verloopt in twee verschillende richtingen. De leidende streng wordt continu gesynthetiseerd, terwijl de achterblijvende streng wordt gesynthetiseerd in korte ketens van 100-200 basenparen lang, de zogenaamde Okazaki-fragmenten. Zodra het grootste deel van de replicatie voltooid is, verwijderen RNase-enzymen de RNA-primers en verbindt DNA-ligase eventuele gaten in de nieuwe streng.

De werklast van het kopi&#xEB;ren van DNA in eukaryoten is verdeeld over meerdere verschillende soorten DNA-polymerase-enzymen. Grote families van DNA-polymerasen in alle organismen worden gecategoriseerd op basis van de gelijkenis van hun eiwitstructuren en aminozuursequenties. De eerste families die ontdekt werden, werden A, B, C en X genoemd, terwijl de families Y en D later werden ge&#xEF;dentificeerd. Familie B-polymerasen in eukaryoten omvat Pol &#x3B1;, dat ook functioneert als een primase bij de replicatievork, en Pol &#x3B4; en &#x3B5;, de enzymen die het meeste werk van DNA-replicatie doen op respectievelijk de leidende en achterblijvende strengen van de matrijs. Omdat eukaryote chromosomen lineair zijn, zijn ze aan de uiteinden vatbaar voor afbraak. Om belangrijke genetische informatie tegen beschadiging te beschermen, bevatten de uiteinden van chromosomen veel niet-coderende herhalingen van sterk geconserveerd G-rijk DNA, de telomeren. Een korte enkelstrengige 3&#x2019; overhang aan elk uiteinde van het chromosoom interageert met gespecialiseerde eiwitten, waardoor het chromosoom in de kern wordt gestabiliseerd. Vanwege de manier waarop de achterblijvende streng wordt gesynthetiseerd, kan een kleine hoeveelheid van het telomere DNA niet bij elke celdeling worden gerepliceerd. Eukaryotic DNA replication copies the genome before a cell divides. In eukaryotes, this process happens in a controlled sequence so each new cell can receive genetic information.

Replication begins at specific sites on the DNA called origins. The DNA is opened, and each original strand serves as a template, meaning it guides the building of a new complementary strand. This sets up two DNA molecules from one original molecule.

The process then continues as enzymes add matching nucleotides to each exposed strand. These new strands are assembled in a way that preserves the DNA sequence. The result is accurate duplication of the genetic material needed for cell division.
