Non-nuclear Inheritance

- [Verteller] In eukaryote cellen, reproduceren mitochondriën en plastiden, een categorie van plantencel organellen die chloroplasten omvat, zich zelfstandig in het cytoplasma en ze hebben elk hun eigen set genen die hun reproductie en functie sturen. Mitochondriaal en plastide DNA heeft de vorm van kleine cirkelvormige DNA-moleculen en wordt non-nucleair of cytoplasmatisch DNA genoemd. Elke organel bevat veel identieke kopieën van zijn DNA.

In tegenstelling tot de kern, waar er doorgaans slechts twee exemplaren van elk chromosoom zijn. Verder worden tijdens de celdeling de organellen willekeurig gescheiden in nieuwe dochtercellen, zodat het non-nucleaire DNA dat wordt doorgegeven geen vast patroon volgt. Tijdens de voortplanting bij dieren en de meeste planten komen het cytoplasma en de zygote uit het ei.

Dus, terwijl beide ouders in gelijke mate bijdragen aan het nucleaire DNA, wordt het non-nucleaire DNA alleen doorgegeven door de moeder. Dit betekent dat bepaalde eigenschappen zoals stofwisselingsstoornissen voortkomend uit mutaties in de mitochondriën op een matrilineaire manier kunnen worden getraceerd. Daarnaast worden sommige fysieke eigenschappen van planten, zoals de bladkleur, geregeld door non-nucleaire genen en worden ze ook door slechts één ouder doorgegeven.

Het meeste DNA bevindt zich in de kern van een cel. Sommige organellen in het celcytoplasma— zoals chloroplasten en mitochondriën—hebben echter ook hun eigen DNA. Deze organellen repliceren hun DNA onafhankelijk van het nucleaire DNA van de cel waarin ze zich bevinden. Niet-nucleaire overerving beschrijft de overerving van genen van andere structuren dan de kern.

Mitochondria zijn aanwezig in zowel planten- als dierlijke cellen. Ze worden beschouwd als de ‘krachtcentrales’. van eukaryote cellen omdat ze glucose afbreken om energie te vormen die de cellulaire activiteit stimuleert. Mitochondriaal DNA bestaat uit ongeveer 37 genen, en veel daarvan dragen bij aan dit proces, dat oxidatieve fosforylatie wordt genoemd.

Chloroplasten worden aangetroffen in planten en algen en zijn de plaatsen van fotosynthese. Door fotosynthese kunnen deze organismen glucose uit zonlicht produceren. Chloroplast-DNA bestaat uit ongeveer 100 genen, waarvan er vele betrokken zijn bij fotosynthese.

In tegenstelling tot chromosomaal DNA in de kern, houden chloroplast- en mitochondriaal DNA zich niet aan de Mendeliaanse veronderstelling dat de helft van het genetische materiaal van een organisme van elke ouder afkomstig is. Dit komt omdat spermacellen tijdens de bevruchting over het algemeen geen mitochondriaal of chloroplast-DNA aan zygoten bijdragen.

Terwijl een spermacel voornamelijk één haploïde set nucleaire chromosomen aan de zygote bijdraagt, draagt een eicel naast zijn nucleaire chromosomen ook zijn organellen bij. Zygoten (en chloroplasten in plantencellen) ontvangen mitochondriën en chloroplasten doorgaans uitsluitend uit de eicel; dit wordt moederlijke erfenis genoemd. Maternale overerving is een vorm van niet-nucleaire of extranucleaire overerving.

Waarom hebben mitochondriën en chloroplasten hun eigen DNA? De heersende verklaring is de endosymbiotische theorie. De endosymbiotische theorie stelt dat mitochondriën en chloroplasten ooit onafhankelijke prokaryoten waren. Cytoplasmic inheritance explains how some traits are passed through cell structures outside the nucleus. These traits come from organelles such as mitochondria and chloroplasts. They do not follow the usual nuclear pattern of inheritance.

Mitochondria and chloroplasts each contain their own DNA. Because of this, they can carry genetic information that affects cell function and certain visible traits. In high school biology, this topic helps explain why not all inherited traits come from chromosomes in the nucleus.

Cytoplasmic inheritance is often linked to the mother because the egg cell provides most of the cytoplasm to the embryo. That is why some traits appear to pass through the maternal line. Understanding this pattern is important for studying organelle genetics and unusual inheritance patterns.
