Mechanisms of Membrane-bending

A membrane has a positive curvature when it bends towards the inner or cytoplasmic side. In contrast, a membrane has a negative curvature if it bends towards the outer or extracellular side.
There are multiple mechanisms that can cause membrane bending.
Lipid-induced membrane bending is due to the asymmetric aggregation of specific lipids in the membrane.
Lipids with large head groups induce positive membrane curvature, whereas lipids with small head groups produce a negative membrane curvature.
Protein scaffolding is another mechanism where proteins either deform the membrane or stabilize an already deformed membrane.
In a different mechanism, hydrophobic protein domains are reversibly inserted in one layer of the membrane to increase its area compared to the other layer. This asymmetry forces the membrane to bend.
Finally, the push and pull forces generated by the assembly and disassembly of the cytoskeleton and its associated motor proteins can also bend the membrane to form projections, such as filopodia in epithelial cells.

Membraanbuiging kan optreden als gevolg van intrinsieke veranderingen in de lipidensamenstelling of extrinsieke associatie met verschillende eiwitten. De eiwitten die betrokken zijn bij het creëren van membraanasymmetrie of clustering van specifieke lipiden spelen een essentiële rol bij door lipiden geïnduceerde membraanbuiging. De vorm van integrale eiwitten kan ook de buiging van het membraan beïnvloeden. Conische of omgekeerd-conisch gevormde transmembraaneiwitten veroorzaken bijvoorbeeld buiging in het membraan. Het inbrengen van amfipathische eiwitten, zoals epsine, in een van de lagen resulteert ook in het buigen van het membraan.

De gebogen eiwitten zoals Bin/amphiphysin/Rvs of BAR-dimeer eiwitten creëren een positieve membraankromming, terwijl het omgekeerde BAR- of I-BAR-eiwit een negatieve kromming in het membraan induceert. BAR-eiwitten brengen ook hun hydrofobe domein in het membraan in, wat de kromming van het membraan verder vergroot. Naast de gevestigde mechanismen is eiwitophoping een recent ontdekt mechanisme voor membraanbuiging, waarbij de laterale druk die wordt gegenereerd door eiwit-eiwitinteracties de membraanbuiging aandrijft. Cell membranes can bend into many shapes because they are flexible and made of a fluid mosaic. That bending can be temporary, as in vesicles, or long-lasting, as in microvilli. Cells control the size, place, and time of membrane curvature.

Membrane bending can start inside the membrane itself or from proteins attaching to it. Changes in lipid makeup can create curvature, and proteins can help by making the membrane asymmetric or by clustering certain lipids. The shape of integral proteins also matters. Conical or inverse-conical transmembrane proteins can push the membrane to bend.

Some proteins bend membranes by inserting part of themselves into one layer of the membrane. Amphipathic proteins, such as epsin, do this and promote curvature. BAR proteins, short for Bin/amphiphysin/Rvs proteins, create positive curvature, while I-BAR proteins create negative curvature. BAR proteins can also add to bending by placing a hydrophobic domain into the membrane.

Protein crowding is another way membranes bend. In this case, proteins packed close together create lateral pressure, and that pressure drives curvature. The cytoskeleton also helps shape membranes. Its forces are important for the form of organelles such as the endoplasmic reticulum and Golgi apparatus.

The folded membranes of the endoplasmic reticulum, Golgi apparatus, and mitochondria are important for their function. Membrane bending also supports membrane scission, membrane fusion, and the activation of enzymes that sense curvature. Changes in genes for membrane-bending proteins, including myotubularin, amphiphysin-2, and dynamin-2, can cause centronuclear myopathies, a group of neuromuscular disorders.
