Cell interactions are physical points of contact between adjacent cells. They help hold the cells together and allow transmembrane communication, vital to the coordinated functioning of tissues.
Stable cell interactions are found in tissues, such as the endothelium, where they support tissue organization. Conversely, weaker, transient interactions help recruit free-moving cells, like immune cells.
These cell interactions materialize through special cell-surface proteins, collectively called cell adhesion molecules or CAMs .
Of the four major families of CAMs, the integrin family is involved in a cell&#8217;s interaction with the extracellular matrix, whereas the other three families are involved in cell-cell interactions.
First are the cadherins, which form stable interactions by binding cadherins expressed on other cells.
Second are the selectins. They bind specific carbohydrates on the surface of other cells and form transient interactions.
And finally, the immunoglobulin-like proteins belonging to the immunoglobulin superfamily also form transient interactions by binding other similar proteins and integrins on other cells.

Celadhesiemoleculen (CAM's) zijn cruciaal voor meercelligheid en het gecoördineerde functioneren van weefsels en orgaansystemen. Ze maken fysieke interacties tussen cellen mogelijk en zorgen voor mechanische sterkte aan weefsels. Ze functioneren ook als receptoren voor signaaloverdracht door het plasmamembraan. De CAM's worden grofweg ingedeeld in vier families: integrinen, cadherinen, selectinen en immunoglobuline-achtige CAM's (IgCAM's).

CAM-families

De Integrine-familie van eiwitten is voornamelijk betrokken bij de interactie van een cel met de omringende matrix. Sommige integrinen kunnen echter CAM's op het oppervlak van een andere cel binden en deelnemen aan directe cel-celinteracties. De integrinen op immuuncellen binden bijvoorbeeld IgCAM's die tot expressie worden gebracht op het vasculaire endotheel.

Cadherinen zijn een superfamilie van calciumafhankelijke glycoproteïnen die voornamelijk betrokken zijn bij het tot stand brengen van sterke celadhesies. Ze vormen de hechtingsverbindingen en desmosomen in weefsels, zoals het epitheel.

Selectinen en IgCAM's zijn betrokken bij voorbijgaande celinteracties en nemen deel aan het sturen van cellen naar doelplaatsen. Ze helpen bijvoorbeeld bij de selectieve rekrutering van lymfocyten naar de secundaire lymfoïde organen.

CAM en meercelligheid

CAM's worden aangetroffen in vrijwel alle meercellige organismen - van sponzen en eenvoudige nematoden tot complexe ongewervelde dieren en gewervelde dieren. De complexiteit van de cellulaire interacties, en dus van de CAM's, neemt toe met de complexiteit van de organismen. Terwijl de fruitvlieg D.melanogaster bijvoorbeeld ongeveer 500 genen heeft die betrokken zijn bij celadhesie, hebben complexe gewervelde dieren zoals zoogdieren meer dan duizend genen die coderen voor verschillende soorten CAM's.

Cell adhesion molecules help cells stick together and work as part of a tissue. They give multicellular organisms physical strength and also help cells send signals across the plasma membrane. These proteins are important for coordinated activity in tissues and organ systems.

CAMs are grouped into four main families: integrins, cadherins, selectins, and immunoglobulin-like CAMs, or IgCAMs. Each family supports cell interactions in a different way. Some CAMs help cells attach firmly, while others help cells make short-lived contacts and move to the right place.

Integrins mainly help a cell interact with the surrounding matrix, which is the material around cells. Some integrins can also bind CAMs on another cell and take part in direct cell-cell contact. For example, integrins on immune cells bind IgCAMs on the vascular endothelium, the inner lining of blood vessels.

Cadherins are calcium-dependent glycoproteins. They are known for strong cell adhesion and help form adherens junctions and desmosomes in tissues such as epithelium. Selectins and IgCAMs usually support transient cell interactions. They also help guide cells to target sites, such as the selective recruitment of lymphocytes to secondary lymphoid organs.

CAMs are found in nearly all multicellular organisms, from sponges and simple nematodes to complex invertebrates and vertebrates. As organisms become more complex, their cell interactions and CAM systems also become more complex. For example, D. melanogaster has about 500 genes involved in cell adhesion, while mammals have over a thousand genes that code for different CAMs.
