The cell cycle refers to the sequence of events throughout a cell&#39;s typical life, including growth, DNA replication, sometimes more growth, and division.
For sexually reproducing organisms, life begins as a zygote, a fertilized egg. Over time, that original single cell grows and divides in a controlled manner to produce a multicellular complex individual.
Cells continue this process, especially to maintain and repair tissues during the progression of aging.
In contrast, single-celled organisms like bacteria rely on cell division to simply reproduce via binary fission.
Unfortunately, when cell division is left unchecked, cancer results, with a loss of control over genomic sequences that regulate the normal process.

De celcyclus verwijst naar de opeenvolging van gebeurtenissen die plaatsvinden gedurende het leven van een typische cel. In eukaryote cellen kent de somatische celcyclus twee fasen: de interfase en de mitotische fase. Tijdens de interfase groeit de cel, voert zijn fundamentele metabolische functies uit, kopieert zijn DNA en bereidt zich voor op mitotische celdeling. Vervolgens verdeelt de cel tijdens mitose en cytokinese respectievelijk zijn nucleaire en cytoplasmatische materialen. Dit genereert twee dochtercellen die identiek zijn aan de oorspronkelijke oudercel. De celcyclus is essentieel voor de groei van het organisme, de vervanging van beschadigde cellen en de regeneratie van verouderde cellen. Kanker is het resultaat van ongecontroleerde celdeling veroorzaakt door een genmutatie.

Controlepunten van de celcyclus

Er zijn drie belangrijke controlepunten in de eukaryotische celcyclus. Bij elk controlepunt kan de voortgang naar de volgende celcyclusfase worden gestopt totdat de omstandigheden gunstiger zijn. Het G1-controlepunt is het eerste hiervan, waar de grootte, energie, voedingsstoffen, DNA-kwaliteit en andere externe factoren van een cel worden geëvalueerd. Als de cel als ontoereikend wordt beschouwd, gaat deze niet door naar de S-fase van de interfase. Het G2-controlepunt is het tweede controlepunt. Hier zorgt de cel ervoor dat al het DNA is gerepliceerd en niet wordt beschadigd voordat het de mitose binnengaat. Als er DNA-schade wordt geconstateerd die niet kan worden gerepareerd; de cel kan apoptose of geprogrammeerde celdood ondergaan. Het M- of spilcontrolepunt zorgt ervoor dat alle zusterchromatiden correct aan de spilmicrotubuli op de metafaseplaat zijn bevestigd voordat de cel in de anafase terechtkomt.

Kanker: wanneer de celcyclus misgaat

Celcycluscontrolepunten zorgen ervoor dat gezonde cellen op een gereguleerde manier door de celcyclus bewegen. Kankercellen omzeilen deze controlepunten echter vaak. Elke opeenvolgende ronde van ongecontroleerde celdeling produceert meer beschadigde dochtercellen. Bovendien kunnen kankercellen in het menselijk lichaam zich veel vaker delen dan normale cellen, die slechts ongeveer 40-60 delingsrondes kunnen ondergaan. Kankercellen brengen telomerase tot expressie, een enzym dat de slijtage aan de uiteinden van chromosomen herstelt, wat doorgaans wordt veroorzaakt door celdeling.
