Cellular Differentiation

During cellular differentiation unspecialized cells take on characteristic features, such as shape, size, or membrane protein composition, to perform distinct functions in the body.
A fertilized egg or zygote is highly proliferative and totipotent; that is, it can differentiate to form all cell types.
As the zygote undergoes a series of divisions, the daughter cells lose their totipotency and differentiate to form specialized cells in the embryo, such as muscle cells, neurons, skin epidermis, heart cells, and liver cells.
Though all these body cells contain the same genome, they express certain genes while switching off others. This allows them to synthesize the proteins necessary to carry out tissue-specific functions.

Het begint allemaal met een enkele bevruchte eicel die een scala aan celtypen voortbrengt, zoals zenuwcellen, spiercellen en epitheelcellen, die kenmerkend zijn voor volwassenen. Gedurende de ontwikkeling en volwassenheid zorgt celdifferentiatie ervoor dat cellen hun uiteindelijke morfologie en fysiologie aannemen. Differentiatie is het proces waarbij ongespecialiseerde cellen, gespecialiseerd worden om specifieke functies uit te voeren.

Een zygote is een totipotente stamcel met een zeer hoog differentiatiepotentieel dat alle celtypen kan vormen en een geheel nieuw multicellulair organisme kan produceren. Naarmate de zygote opeenvolgende celverdelingen ondergaat, worden pluripotente stamcellen (PSC's) gegenereerd met een meer beperkt differentiatiepotentieel. PSC's geven aanleiding tot een paar volwassen stamcellen die verder delen en meer terminaal gedifferentieerd worden. Wanneer een cel differentieert, kan deze significante veranderingen ondergaan in grootte, vorm, metabole activiteit en algehele functie.

Vanaf de bevruchte eicel bevatten alle cellen in het lichaam hetzelfde DNA, maar elk celtype "leest" alleen de delen van het DNA die relevant zijn voor zijn functie. Verschillende factoren zoals hormonen of signaalmoleculen zorgen voor celdifferentiatie en schakelen als reactie daarop de expresie van specifieke genen aan en andere uit. Het primaire mechanisme waardoor genen "aan" of "uit" worden gezet, vindt plaats via transcriptiefactoren. Een transcriptiefactor is een klasse eiwitten die binden aan specifieke genen op het DNA-molecuul en hun transcriptie bevorderen of remmen. De expressie van specifieke genen bemiddelt de synthese van noodzakelijke eiwitten die vereist zijn voor een bepaalde cel functie, en helpt bij cel differentiatie.

Cellular differentiation turns unspecialized cells into specialized cells with specific jobs. This process helps a complex organism, such as a human, develop from a single fertilized egg into a body with many cell types. Nerve cells, muscle cells, and epithelial cells are examples of the final cell types that appear during development.

A zygote is the first cell after fertilization. It is a totipotent stem cell, which means it has very high differentiation potential and can form all cell types and even produce a whole new multicellular organism. As the zygote divides, it gives rise to pluripotent stem cells, or PSCs, which have a more limited ability to differentiate. PSCs then produce a few adult stem cells that keep dividing until they become more terminally differentiated.

As cells differentiate, they can change in size, shape, metabolic activity, and overall function. Even though the body’s cells contain the same DNA, each cell type only reads the parts of the DNA that are relevant to its role. This selective reading helps each cell become suited to its own job.

Hormones and signaling molecules can trigger differentiation. In response, specific genes are turned on while others are turned off. Transcription factors, which are proteins that bind to specific genes on DNA, control this switch by promoting or inhibiting transcription. The expression of the right genes leads to the proteins needed for a particular cell function and supports cellular differentiation.
