The noncompartmental approach utilizes various factors, including clearance, bioavailability, and total volume of distribution.
Once a drug is administered, the total clearance of a drug can be determined by dividing the drug dose by the area under the concentration-time curve from zero to infinity as obtained with the trapezoidal method.
Bioavailability, F, can be determined using the drug concentration data after the oral and intravenous administration of a drug.
The total volume of distribution, Vss, is the product of clearance and mean residence time.
Mean residence time can be readily calculated only after a single dose of intravenous administration, which is a limitation of the noncompartmental approach compared to the compartmental approach.
