Chronopharmacokinetics studies the temporal change in drug absorption and elimination, whether cyclical or noncyclical.
Cyclic changes occur over constant intervals, while noncyclic changes extend over time.
Time-dependent pharmacokinetics refers to noncyclical changes in drug rate processes, leading to nonlinear pharmacokinetics.
It may result from changes in biochemistry or alteration in the physiology of organs, possibly impacting drug disposition.
Additionally, it can be due to the autoinduction or autoinhibition of biotransformation enzymes.
Autoinduction occurs when repeated drug doses induce the enzymes responsible for its elimination, increasing drug clearance.
Autoinhibition occurs when metabolites increase in concentration and inhibit the metabolism of the parent drug.
Drugs with time-dependent pharmacokinetics have variable clearance and elimination half-lives.
At certain periods of the day, some anticancer drugs are better tolerated. For instance, the antimetabolite drug fluorouracil, administered in rodents during the morning, was found to be the least toxic.
