The Mantel-Cox log-rank test is a nonparametric statistical method for comparing survival distribution curves between two groups.
It is typically employed in clinical research to evaluate treatment efficacy over time and guide further studies.
For instance, researchers can use this test to determine statistically significant differences between the survival curves of one group undergoing a novel therapy and another undergoing the control treatment.
The Mantel-Cox test calculates differences between observed and expected events across groups without assuming a specific survival time distribution. It is ideal for analyzing censored data in which not all subjects may experience the event of interest, like death or disease relapse.
Its limitation lies in its reliability on the proportional hazards assumption, which posits constant hazard ratios over time and may only sometimes be true. The results of this test could be misleading if the proportional hazard assumption is violated.
This is particularly true for studies with small sample sizes or high censoring rates.
