Deze video beschrijft een techniek van Embryo microinjection om verbindingen in Zebrafish dooierzak te leveren.

Protocol

1. Micro-invoeging van CRISPR-componenten in Zebrafish Embryo's

1. Zet foktanks op de nacht voorafgaand aan het injecteren door het aantal gewenste mannetjes en vrouwtjes (meestal 2 vrouwtjes en 1 of 2 mannetjes) in een kweektank met een scheidingswand op zijn plaats te plaatsen.
2. Bereid een micro-invoegplaat met 1,5% agarose in 1x E3-media (zie tabel met materialen) met 0,01% methyleenblauw (een fungicide) door 35 ml van de gesmolten agarose in een Petrischaal van 10 cm te gieten en leg voorzichtig een plastic mal om wigvormige troggen in de oplossing te maken, waarbij de mal wordt aangetikt om luchtbellen te elimineren.

Resultaten

Figuur 1: Vergelijking van de gezondheid van 24 hpf geïnjecteerde embryo's. Een levend embryo (A) ontwikkeld tot 24 hpf, is gemakkelijk te onderscheiden van een embryo dat de ontwikkeling heeft afgebroken (B). Embryo's die lijken op (B) of die drastisch zijn veranderd in (A), zoals kromming van de wervelkolom of veranderde hoofd- en oogontwikkeling, moeten uit de schaal worden verwijderd
