Deze video demonstreert de productie en zuivering van recombinante hepatitis B-virusdeeltjes, wat veilige, replicatie-deficiënte virale preparaten mogelijk maakt voor het bestuderen van vroege infectiegebeurtenissen en voor high-throughput screeningtoepassingen van antivirale geneesmiddelen.

Protocol

1. Productie van recombinant HBV (hepatitis B-virus) dat het reportereiwit codeert

1. Preparatie van HepG2-cellen
   1. Bereid celkweekmedium voor (Dulbecco's modified Eagle's medium (DMEM) aangevuld met 10% foetaal runderserum (FBS), 100 U/ml penicilline, 100 µg/ml streptomycine en 100 U/ml niet-essentiële aminozuren).
   2. Zaai de dag voor transfectie 4 x 10⁶ HepG2-cellen (Human Hepatocellular Carcinoma G2) uit in een met collageen gecoate schaal van 10 cm in 10 ml kweekmedium. Incubeer de HepG2-cellen bij 37 °C in een bevochtigde 5% CO₂ incubator.
