The catheter of a telemetry device is implanted into the abdominal aorta in order to continuously collect beat-by-beat hemodynamic data from animals pre and post-high thoracic spinal cord transection. A novel JAVA software was employed to analyze hemodynamic parameters as well as frequency and intensity of spontaneous episodes of autonomic dysreflexia.

Spinal cord injury (SCI) is a debilitating neurological condition characterized by somatic and autonomic dysfunctions. In particular, SCI above the mid-thoracic level can lead to a potentially life-threatening hypertensive condition called autonomic dysreflexia (AD) that is often triggered by noxious or non-noxious somatic or visceral stimuli below the level of injury. One of the most common triggers of AD is the distension of pelvic viscera, such as during bladder and bowel distension or evacuation. This protocol presents a novel pattern recognition algorithm developed for a JAVA platform software to study the fluctuations of cardiovascular parameters as well as the number, severity and duration of spontaneously occurring AD events. The software is able to apply a pattern recognition algorithm on hemodynamic data such as systolic blood pressure (SBP) and heart rate (HR) extracted from telemetry recordings of conscious and unrestrained animals before and after thoracic (T3) complete transection. With this software, hemodynamic parameters and episodes of AD are able to be detected and analyzed with minimal experimenter bias.

Autonome dysreflexie (AD) is een levensbedreigende noodsituatie patiënten na acute of chronische ruggenmergletsel (SCI) en cervicale of high-thoracale segmenten en wordt gewoonlijk gekenmerkt door episoden van aanhoudende hypertensie en bradycardie 1. AD wordt voornamelijk veroorzaakt door een verstoring van de dalende spinale routes die gewoonlijk een input van supraspinale centra naar de spinale sympathische preganglionic neuronen die sympathische activiteit en vasculaire tonus 1-4 te regelen. AD episodes worden gekenmerkt door een piek in de systolische bloeddruk (SBP) tot 300 mmHg en indien onbehandeld kan leiden tot convulsies, intracraniël....

Protocol

Mannelijke Wistar (Hsd: WI Wistar) ratten 7 weken oud en een gewicht van 300-350 g werden gebruikt in dit experiment. Alle ratten werden gehandhaafd op een 12 uur licht / donker-cyclus en kreeg standaard laboratorium rat chow en water ad libitum. Alle experimentele procedures in overeenstemming met de gids van de Zorg en gebruik van proefdieren opgesteld door de Canadese Raad over Animal Care en verleende ethiek goedkeuring door de University of British Columbia. Chirurgie en verzorging van dieren werden uitgevoerd volgens standaardprocedures in ons laboratorium (Ramsey et al. 2010) 17.

Telemetrie gebruiken, wordt de arteriële bloeddruk bemonsterd met een frequentie van 1000 Hz continu gedurende 24 uur. Een illustratieve registratie van de arteriële bloeddruk (ABP) middels LabChart is weergegeven in figuur 1B. Het monster ABP werd gevolgd door een halfgeleider druksensor geplaatst in de aorta descendens. Het protocol wordt een gedetailleerde uitvoering van een nieuwe JAVA platform AD detectie software die worden gecombineerd met een telemetrie-inrichting voor een langdurige grondige analyse van ABP in SCI-dieren (Figuur 1B).
