Dit artikel presenteert een methode voor het opzetten van een in vitro psoriasiform cutaan inflammatoir model op transcriptieniveau met behulp van een combinatie van vijf cytokines (IL-17A, IL-22, IL-1α, TNF-α, OSM) op de HaCaT-cellijn.

Samenvatting

Psoriasis is een veel voorkomende chronische inflammatoire huidziekte die wordt gemedieerd door aangeboren en adaptieve immuunsystemen, gekenmerkt door abnormale proliferatie en differentiatie van epidermale keratinocyten en infiltratie van ontstekingscellen. Huidspecifieke keratinocyten zijn belangrijke deelnemers aan aangeboren immuniteit, reageren op immuuncellen en omgevingsstimulatie, en spelen daardoor een belangrijke rol in de immunopathogenese van psoriasis. Hier presenteren we een methode voor het induceren van psoriasiforme keratinocytenontsteking op transcriptieniveau met HaCaT-cellijn met behulp van vijf pro-inflammatoire cytokinecombinaties (M5-combinatie), waaronder IL-17A, IL-22, IL-1α, TNF-α en oncostatine M. De resultaten tonen aan dat door M5-combinatie geïnduceerde HaCaT-cellen verhoogde niveaus van antimicrobiële peptiden (BD2, S100A7, S100A8 en S100A9), chemokines en cytokines (CXCL1, CXCL2, CXCL8, CCL20, IL-1β, IL-6 en IL-18). De mRNA-niveaus van keratinocytendifferentiatiemarkers (Keratin1, Keratin10, Filaggrin en Loricrin) waren neerwaarts gereguleerd, wat consistent was met de transcriptoomgegevens afgeleid van psoriasis-achtige keratinocyten. De hier beschreven methode stelt dus een in vitro psoriasiforme huidontsteking op transcriptieniveau vast en draagt bij aan het onderzoek naar moleculaire pathogenese van psoriasis.

Inleiding

Psoriasis is een veel voorkomende niet-besmettelijke chronische inflammatoire huidziekte die wordt veroorzaakt door een ontregelde immuunrespons, die de keratinocyten aantast die voornamelijk de epidermis1 vormen, gekenmerkt door abnormaal snelle vermenigvuldiging van keratinocyten met hyperkeratose en parakeratose. Psoriasis treft ongeveer 3% van de wilde wereldpopulatie2. De ziektelast wordt verder verhoogd door verschillende comorbiditeiten, waaronder hart- en vaatziekten en het metabool syndroom veroorzaakt door het syndroom3.

Epidermis is samengesteld uit vijf lagen keratinocyten en ondergaan morfologische verandering met differentiatieproces: de laag basaal, stratum spinosum, stratum granulosum, stratum lucidum (te vinden op handpalmen en voetzolen) en stratum corneum hier beschreven van het binnen- tot buitenoppervlak4. Verandering in epidermisdifferentiatie leidt tot een verstoorde huidbarrière, wat belangrijk is voor de pathogenese van huidontstekingsziekten 5,6,7,8. Keratinocyten spelen een cruciale rol bij het behoud van een intacte epidermale barrière om waterverlies te voorkomen en tegen omgevingstriggers zoals blootstelling aan UVB, allergenen en ziekteverwekkers9. Gezonde individuen vertonen een evenwicht tussen proliferatie van basale cellen en afschilfering van het stratum corneum, terwijl meerdere huidziekten, waaronder psoriasis, worden gekenmerkt door een onevenwichtigheid van dit complexe mechanisme10.

Keratinocyten vormen niet alleen een barrièrefunctie, maar zijn ook een cruciaal onderdeel van het immuunsysteem van de huid. In de immunopathogenese van psoriasis leidt activering van huidbewoner Type 1 helper T-cellen (Th1) en Type 17 helper T-cellen (Th17) tot een verhoogde productie van respectievelijk IFN-γ en IL-17A. Deze cytokinen induceren een verhoogde synthese van chemokines (CCL20, CXCL1/2/8/9/10/11), antimicrobiële peptiden (BD2, LL37, S100A7/8/9/12) en andere ontstekingsfactoren (TNF-α, IL-6, IFN-β) in keratinocyten, wat leidt tot de rekrutering van meer Th1, Th17 en neutrofielen in de huid, waardoor de IL-17/IL23-as11 verder wordt versterkt. De overspraak tussen keratinocyten en immuuncellen is verantwoordelijk voor de inductie en het onderhoud van psoriasis11.

Complexe cytokinenetwerken zijn beschreven bij psoriasis en de centrale rol van pro-inflammatoire cytokinen (zoals IL-23, IL-22, IL-17, IL-1α, oncostatine M (OSM) en TNF-α) geproduceerd door infiltratie van immuuncellen is benadrukt12,13. Eerdere studies hebben inderdaad aangetoond dat verhoogde niveaus van IL-17A, IL-22, IL-1α, TNF-α en OSM een profiel van psoriasiform induceerden op normale menselijke epidermale keratinocyten in vitro14.

Onsterfelijke keratinocytcellijnen (HaCaT) die gemakkelijker kunnen worden verkregen en gekweekt dan primaire keratinocyten met een betere reproduceerbaarheid, zijn veel gebruikt voor de studie van psoriasis 15,16,17,18,19,20. Anders dan humaan papillomavirus16 E6/E7 getransformeerde HEK001- en KerTr-cellen, is de HaCaT-cellijn in staat om differentiatiegenproducten tot expressie te brengen, waaronder keratine1 (KRT1), keratine10 (KRT10), loricrine en filaggrine 20,21,22, waardoor een veelbelovend hulpmiddel wordt geboden dat vergelijkbaar is met primaire keratinocyten om de regulatie van keratinisatie en pro-inflammatoire te bestuderen.

KRT5/14 is het belangrijkste type I-type II keratinepaar dat tot expressie komt in proliferatieve basale keratinocyten, terwijl gedifferentieerde keratinocyten in de suprabasale lagen KRT5/14 downreguleren en KRT1/10 tot expressie brengen als het belangrijkste keratinepaar23. Bij vergelijking van psoriasislaesie met een gezonde huid omvatten de veranderingen in keratine-expressie dalingen in KRT1/1024,25 en verhogingen in KRT5/14 in de psoriatische epidermis26, gekenmerkt door hyperproliferatie en parakeratose27. Loricrine is een terminaal differentiërend structureel eiwit dat meer dan 70% van de verhoornde envelop omvat, draagt bij aan de beschermende barrièrefunctie van de hoornlaag28, dons gereguleerd in de huid van psoriasispatiënten29. Filaggrine komt tot expressie in de laatste stadia van keratinocytendifferentiatie en is betrokken bij de aggregatie van een steigerachtige verhoornde envelop30, verminderde expressie in de huid van psoriasislaesies29.

Over het algemeen was ons doel om een inflammatoir keratinocytenmodel in HaCaT-cellen te genereren met behulp van een cytokinecombinatie die in staat zal zijn om synergetisch enkele kenmerken van huidlaesies van psoriasis samen te vatten, waaronder het initiëren van een immuunrespons, proliferatie en differentiatie van keratinocyten.

Protocol

Voer stap 1 tot en met 3 uit onder steriele toestand. Al het kweekmedium bevatte 0,1 mg/ml penicilline en streptomycine.

1. Voorbereiding van de cellen

  1. Zaai 1 x 106 HaCaT-cellen in 10 ml Dulbecco's Modified Eagle Medium (DMEM) aangevuld met 10% foetaal runderserum (FBS) in 100 mm celkweekschaaltje. Incubeer de kweekschaal gedurende 2 dagen bij 37 °C in een bevochtigde 5% CO2 -incubator.
  2. Wanneer de cultuur ongeveer 80% samenvloeiing bereikt, verwijdert u het medium voorzichtig uit de kweekschaal en wast u de cellen met 5 ml 1x fosfaatgebufferde zoutoplossing (PBS). Schud de kweekschaal voorzichtig met de hand.
  3. Verwijder PBS en voeg 2 ml 0,25% trypsine-EDTA-oplossing toe aan de cellen. Schud de kweekschaal voorzichtig om de oplossing de monolaag cellen volledig te laten bedekken.
  4. Incubeer de cellen bij 37 °C in een bevochtigde 5 % CO2 -incubator gedurende 5 minuten totdat de cellen zichtbaar zijn losgemaakt van het oppervlak van de kweekschaal onder fasecontrastmicroscopie.
    OPMERKING: De morfologie van cellen moet er rond uitzien. Als de cellen niet goed losgemaakt zijn, incubeer dan gedurende een extra periode van 5 minuten samen met handmatig roeren.
  5. Zodra de cellen zijn losgemaakt, voegt u 5 ml DMEM plus 10% FBS toe om trypsine te inactiveren en verzamelt u de celsuspensie in een centrifugebuis van 50 ml met behulp van een pipet.
  6. Centrifugeer de buis met de celsuspensie op 180 x g gedurende 5 min. Decanteer de bovengrondse.
  7. Voeg 10 ml DMEM plus 10% FBS-medium toe aan de pellet en pipetteer het medium voorzichtig op en neer om de cellen in suspensie te brengen.

2. Celzaaien in de plaat met 6 putjes

  1. Zaai de cellen met een dichtheid van 2,0 x 105 cellen/putje in een kweekplaat met 6 putjes.
  2. Incubeer de 6-putjes kweekplaat bij 37 °C in een bevochtigde 5% CO2 -incubator een nacht voordat u de M5-combinatiestimulatie uitvoert om de cellen aan de plaat te laten hechten.

3. M5-stimulatie van HaCaT-cellen

  1. Bereid een M5-cytokinecombinatiemengmedium dat 10 ng/ml recombinant IL-17A-, IL-22-, IL-1α-, TNF-α- en oncostatine-M-eiwit in DMEM bevat dat 2% FBS bevat.
    OPMERKING: De M5-combinatie bestond uit recombinatie van IL-17A, IL-22, IL-1α, TNF-α en oncostatine M. De synergetische werking van de M5-combinatie recapituleert enkele kenmerken van psoriasis, zoals opregulatie van chemokinen en de productie van antimicrobiële peptiden31.
  2. Verwijder na een nacht celkweek de supernatant en pipetteer 2 ml M5-combinatiemengmedium in elk putje.
  3. Ga door met het kweken van de cellen; cellysaten werden verzameld om 24 uur voor mRNA-kwantificering (stap 4) of kweeksupernatans werden verzameld om 48-72 uur voor de bepaling van cytokineniveaus door middel van ELISA-assay 32,33,34,35,36,37 (stap 6).

4. Oogst mRNA van M5 gestimuleerde HaCaT-cellen

  1. Zuig de M5 combinatiemix medium op en was eenmaal met 1-2 ml koude PBS.
  2. Aspireer PBS en voeg 1 ml in de handel verkrijgbare guanidiumisothiocyanaatoplossing rechtstreeks toe aan de kweekschaal om de cellen te lyseren.
  3. Pipetteer het lysaat meerdere keren op en neer om het te homogeniseren en breng het cellysaat over in een microcentrifugebuis van 1,5 ml. Laat 5 minuten op kamertemperatuur staan.
  4. Voeg 200 μL chloroform toe, schud de tube krachtig gedurende ongeveer 20 s en incubeer het monster gedurende 5 minuten bij kamertemperatuur.
  5. Centrifugeer bij 12.000 x g gedurende 10 minuten bij 4 °C.
  6. Breng de waterige fase over in een verse microcentrifugebuis van 1,5 ml.
  7. Voeg 500 μL isopropanol toe aan de waterige fase en meng voorzichtig. Laat 5 minuten op kamertemperatuur staan.
  8. Centrifugeren bij 10.000 x g gedurende 15 minuten bij 4 °C.
  9. Aspireer isopropanol op en voeg 1 ml 75% ethanol toe. Was de korrel één keer.
  10. Centrifugeren bij 7.500 x g gedurende 5 minuten bij 4 °C. Giet de ethanol af en laat de pellets aan de lucht drogen.
  11. Voeg 30 μL DEPC behandeld water toe aan de RNA-pellet. Bereid je voor op RT-PCR-detectie.

5. Analyse van mRNA-expressie door Real-Time PCR

  1. Voer cDNA-synthese uit met 1 μg totaal RNA met behulp van een in de handel verkrijgbare kit volgens de instructies van de fabrikant.
    OPMERKING: 1 μg totaal RNA voor SYBR Green RT-PCR-test.
  2. Bereid het PCR-reactiemengsel voor. 12,5 μl SYBR premix EX Taq II, 1 μl PCR Forward Primer (10 μM), 1 μl PCR Reverse Primer (10 μM), 2 μg cDNA en 8,5 μl dH2O. Het uiteindelijke volume is 25 μL per reactie.
    OPMERKING: Lijst met primersequenties van genen die zijn gebruikt voor RT-PCR-analyse in deze studie wordt weergegeven in tabel 1.
  3. Incubeer in een thermocycler. De cycluscondities omvatten een denatureringsstap bij 95 °C gedurende 30 s, 40 cycli van 95 °C gedurende 5 s, 60 °C gedurende 30 s, 72°C gedurende 20 s en een analyse van de smeltcurve.
  4. Analyseer RT-PCR-experimentgegevens door relatieve verschillen in genexpressie te berekenen op basis van Ct-waarden (drempelcyclus) met behulp van 2-ΔΔCT-vergelijkingen. Gebruik β-actine als interne controle voor RT-PCR relatieve kwantitatieve genexpressiestandaardisatie.

6. Oogsten van celcultuur supernatans voor ELISA

  1. Pipetteer elk celkweekmedium in een microcentrifugebuis van 1,5 ml.
  2. Centrifugeer bij 1500 x g gedurende 10 minuten bij 4 °C.
  3. Aliquot het bovenstaande en onmiddellijk bewaren bij -80 °C.

7. Analyse van de expressie van cytokines door ELISA

  1. Volg de instructies van de fabrikant om alle reagentia, werkstandaard en monsters voor te bereiden. Een drievoudige seriële verdunning van de standaard met monsterverdunningsmiddel variërend van 2.000 tot 2,74 pg/ml. Verdun de monsters 1:2 met monsterverdunningsmiddel.
  2. Voeg 100 μL standaard en monster per putje toe. Dek de putjes af met een deksel en sluit de plaat af met paraffinefolie. Incubeer gedurende 2,5 uur bij kamertemperatuur.
  3. Gooi de oplossing na 2,5 uur weg. Voeg 300 μL wasbuffer toe aan elk putje van de plaat gedurende 3 minuten en zuig op. Herhaal het proces nog vier keer. Keer na de laatste wasbeurt de plaat om en tik op absorberend papier om de resterende vloeistof te verwijderen.
  4. Voeg 100 μL van de detectie-antilichaamoplossing toe aan elk putje. Incubeer de plaat op een shaker gedurende 2 uur bij 37 °C.
    OPMERKING: In elke ELISA-kit wordt een biotinyleerde detectie-antilichaamoplossing bereid.
  5. Zuig elk putje van de plaat op en was het vijf keer met 300 μL wasbuffer. Keer na de laatste wasbeurt de plaat om en tik op absorberend papier om de resterende vloeistof te verwijderen.
  6. Voeg 100 μL HRP-Streptavidin-conjugaat toe aan elk putje. Incubeer gedurende 45 minuten bij kamertemperatuur en schud zachtjes.
  7. Gooi de oplossing weg door na de incubatie met een pipet op te zuigen. Was elk putje van de plaat vijf keer met 300 μL wasbuffer. Keer na de laatste wasbeurt de plaat om en tik op het absorberende papier om de resterende vloeistof te verwijderen.
  8. Voer de detectie uit door 100 μL TMB-substraat aan elk putje toe te voegen. Incubeer gedurende 30 minuten bij 37 °C in het donker.
  9. Voeg 50 μL stopoplossing toe, wanneer de kleur zich ontwikkelt, aan elk putje. Tik zachtjes op de plaat om een grondige menging te garanderen. Onmiddellijk aflezen bij 450 nm.

Resultaten

M5-combinatiestimulatie induceerde ontstekingsreactie van HaCaT-cellen.
HaCaT-cellen werden gedurende 24 uur gestimuleerd met of zonder combinatie van M5-cytokines. mRNA-expressie van psoriasis-gerelateerde genen, die betrokken zijn bij de regulatie van de immuun- en inflammatoire chemokines en antimicrobiële peptiden, werden geëvalueerd. Neutrofiele chemokines CXCL138, CXCL23...

Discussie

Hierin wordt een methode beschreven waarbij gebruik wordt gemaakt van een combinatie van vijf cytokines (IL-17A, IL-22, IL-1α, TNF-α, OSM) in de HaCaT-cellijn om een in vitro psoriasiform huidontstekingsprofiel op transcriptieniveau vast te stellen.
