Dit protocol presenteert een op microcontrollers gebaseerde gedragsbox om prepulsinhibitie (PPI) te beoordelen door versnellingsgegevens te verzamelen van een sensor onder de box. Deze gegevens evalueren sensorische poorttekorten bij sociaal geïsoleerde ratten, met een toegevoegde methode voor het synchroniseren van gegevens met neuronale activiteit om neurofysiologische studies naar gedragsstoornissen vooruit te helpen.

Abstract

Nood in het vroege leven wordt erkend als een mogelijke voorloper van neurologische ontwikkelingsstoornissen. Een benadering om deze aandoening in diermodellen te onderzoeken, is sociaal isolement na het spenen. Het is aangetoond dat dit leed in het vroege leven bij knaagdieren leidt tot neurale veranderingen die vergelijkbaar zijn met die waargenomen bij patiënten met schizofrenie, waaronder een verminderd volume van de prefrontale cortex, verminderde dichtheid van de dendritische wervelkolom en verhoogde dopaminerge activiteit in het ventrale tegmentale gebied. Het prepulsinhibitieparadigma (PPI) is een gevestigde methode voor het beoordelen van sensorische poorttekorten in zowel menselijke als diermodellen. PPI verwijst naar de onderdrukking van de schrikreflex naar een sterkere akoestische stimulus (bijv. 120 dB puls) wanneer deze wordt voorafgegaan door een zwakkere akoestische pre-stimulus (bijv. 65 dB puls). Dit fenomeen wordt vaak verminderd bij psychiatrische patiënten en diermodellen van psychiatrische stoornissen. Dit artikel introduceert een protocol om een gedragsbox te bouwen voor PPI-studies, waardoor het gebruik van een toegankelijk, kosteneffectief diermodel van schizofrenie mogelijk wordt en de synchronisatie van de gedragsbox met neuronale opnames van het model (bijv. elektrofysiologische opnames). Deze op maat gemaakte gedragsbox detecteerde effectief schrikreacties en faciliteerde PPI-beoordeling met gelijktijdige neuronale registratie bij zich vrij gedragende knaagdieren die werden onderworpen aan isolatie na het spenen.

Personen met de diagnose schizofrenie (SCZ) vertonen vaak sensomotorische stoornissen. Verminderde respons op sensorische stimulatie en moeite met het onderscheiden van relevante informatie uit diverse sensorische inputs wordt vaak gemeld bij die patiënten 1,2. Voor gezonde personen vermindert het toedienen van een zwakkere stimulus vóór een sterkere de gebruikelijke schrikreflex naar de sterkere stimulus3. Dit fenomeen staat bekend als prepulsinhibitie (PPI) en is gebruikt om sensorische poorttekorten te onderzoeken bij patiënten ....

Protocol

Dierproeven zijn uitgevoerd in overeenstemming met de Leidraad voor de verzorging en het gebruik van proefdieren. Dit project werd goedgekeurd door het Ethisch Comité van het CEUA Santos Dumont Institute (erkenningsnummer 04/2016). Voor PPI-testen werden 35 Wistar-ratten (3 maanden oud, 290-490 g; 19 mannetjes en 16 vrouwtjes) individueel geëvalueerd in de PPI-box. Alle 35 ratten ondergingen een protocol van isolatie na het spenen, waarbij ze individueel werden gehuisvest onder een lichtcyclus van 12/12 uur bij een temperatuur van 23 °C en een luchtvochtigheid van 70% in een gecontroleerde faciliteit met ad libitum toegang to....

Results

Prepulsremming toonde geen verschil voor puls 1 en 3 in vergelijking met puls 5 (alleen 120dB), maar toonde een statistisch verschil voor puls 2 en 4 (p < 0,05) (Figuur 6).

Dit protocol presenteerde een doe-het-zelf-gedragsbox om prepulsinhibitietaken te bestuderen bij zich vrij gedragende Wistar-ratten tijdens het registreren van hersengegevens (in dit voorbeeld lokaal veldpotentieel en piekactiviteit). De doos kan worden gebouwd en aangepast met componenten die vaak worden aangetroffen in neurowetenschappelijke laboratoria, waardoor hij toegankelijk en goedkoop is.
