Bone Resorption Assessment

Bone resorption assessment is the measurement of how rapidly and extensively bone tissue is degraded, providing insight into skeletal remodeling and the performance of engineered bone materials. The process is driven primarily by osteoclasts, which attach to mineralized surfaces, create an acidic microenvironment that dissolves mineral, and release enzymes that break down the organic matrix. In bioengineering, researchers assess resorption using bone-cell cultures, mineralized substrates, imaging, mass loss, or biochemical markers released during matrix degradation. These measurements help evaluate biomaterial biocompatibility, scaffold design, implant stability, and the balance between bone degradation and formation in regenerative strategies.

In this video, we describe an in vitro osteoclastic resorption assay on calcium phosphate-coated cell culture plates cultured with human peripheral blood mononuclear cell-derived osteoclasts. The osteoclasts form a characteristic actin ring on the calcium phosphate-coated matrix of the cell culture plate and degrade the matrix within the actin ring region to release calcium and phosphate ions, resulting in the formation of distinct resorption pits within the degraded matrix.

An Improved Mechanical Testing Method to Assess Bone-implant Anchorage

An improved method to mechanically test bone anchorage to candidate implant surfaces is presented. This method allows for alignment of the disruption force exactly perpendicular, or parallel, to the plane of the implant surface, and provides an accurate means to direct the disruption forces to an exact peri-implant region.
