Chip Loading Procedure

The chip loading procedure is a technique for introducing biological samples, reagents, or cells into the defined channels and chambers of a microfluidic chip before analysis. It typically involves priming the device, controlling pressure or flow, and dispensing a measured volume through an inlet while preventing air bubbles, leaks, and cross-contamination. Proper loading helps distribute material consistently and preserves the intended reaction or separation conditions within the chip. In biological research, this step supports applications such as cell handling, molecular assays, droplet generation, and single-cell analysis, where loading accuracy can influence experimental reproducibility and data quality.

Accessible Silicone Chip-to-Membrane Sealing Procedure for Flexible, Reliable Bonding

A novel sealing protocol to produce water-tight silicone-based seals for membranes of Polydimethylsiloxane (PDMS) chip devices. Intended for labs setting-up chip models and small-scale chip platform development. We demonstrate the protocol using plastic and native tissue membranes. This procedure only requires PDMS, toluene, mold, membrane, and a vacuum chamber.

Procedure for the Development of Multi-depth Circular Cross-sectional Endothelialized Microchannels-on-a-chip

A microchannels-on-a-chip platform was developed by the combination of photolithographic reflowable photoresist technique, soft lithography, and microfluidics. The endothelialized microchannels platform mimics the three-dimensional (3D) geometry of in vivo microvessels, runs under controlled continuous perfusion flow, allows for high-quality and real-time imaging and can be applied for microvascular research.

A Semiautomated ChIP-Seq Procedure for Large-scale Epigenetic Studies

This paper describes a semiautomated, microscaled ChIP-Seq protocol of DNA regions associated with a specific histone modification (H3K27ac) using a ChIP liquid-handler platform, followed by library preparation using tagmentation. The procedure includes control assessment for quality and quantity purposes and can be adopted to other histone modifications or transcription factors.

Chip-Based Digital PCR to Detect Rare Transcript Variants Using a Nanofluidic Chip

This video demonstrates chip-based digital PCR — a variation of the digital PCR technique that is useful in detecting rare transcript variants. The PCR reaction is partitioned into the chambers of a nanofluidic chip, each of which acts as an independent reaction. The detection of fluorescence signals from the chambers with amplified targets confirms the presence of rare transcript variants in the sample.

Lensless Fluorescent Microscopy on a Chip

A lensless on-chip fluorescent microscopy platform is demonstrated that can image fluorescent objects over an ultra-wide field-of-view of e.g., >0.6-8 cm2 with <4μm resolution using a compressive sampling based decoding algorithm. Such a compact and wide-field fluorescent on-chip imaging modality could be valuable for high-throughput cytometry, rare-cell research and microarray-analysis.
