Fluorescence Intensity Fluctuations

Fluorescence intensity fluctuations are temporal changes in the brightness emitted by fluorescent molecules, providing information about molecular behavior that is not captured by a single intensity measurement. These variations arise when molecules diffuse in and out of an observation volume, change conformation or concentration, or switch between fluorescent and nonfluorescent states through photophysical processes such as blinking and bleaching. In biological techniques, analyzing fluctuation amplitude and timescales supports fluorescence correlation spectroscopy and related approaches for measuring diffusion, molecular interactions, transport, and reaction kinetics. This information helps researchers characterize cellular organization and dynamic processes at microscopic scales.

Fluorescence Fluctuation Spectroscopy to Study Protein Homo-Oligomerization

This video demonstrates the technique of fluorescence fluctuation spectroscopy (FFS) to study protein homo-oligomerization. Fluorescently-labeled proteins in a sample are dimerized using an agent when studying protein oligomerization using FFS. Using a confocal microscope, as the proteins move in and out of the small observation volume, the analysis of the fluctuations in the brightness of the fluorescent molecules is performed to determine the oligomeric state of the proteins.
