Light Membrane Fraction

The light membrane fraction is a low-density cellular fraction enriched in small membrane-bound structures and isolated from cell or tissue lysates during biochemical fractionation. It is typically separated by differential or density-gradient centrifugation, in which membranes partition according to their size, density, and buoyant behavior while heavier organelles sediment under stronger centrifugal force. Researchers use this fraction to examine membrane proteins, vesicles, intracellular trafficking, and signaling pathways, often alongside marker proteins that identify its cellular components. Studying the light membrane fraction helps link membrane composition and distribution to processes such as secretion, endocytosis, organelle communication, and disease-related changes in cell organization.

2025

This protocol separates insoluble cell wall and membrane proteins into simple fractions (1-5 proteins) using preparative isoelectric focusing (IEF) based on isoelectric point, followed by separation by molecular weight. The resulting fractions can be used directly for immunological and proteomic analysis without further purification.

Here we describe a robust method for the fractionation of plant plasma membranes into detergent resistant and detergent soluble membranes based on a mixture of unlabeled and in vivo fully 15N labeled Arabidopsis thaliana cell cultures. The procedure is applied for comparative proteomic studies to understand signaling processes.
