Membrane Potential Dye

Membrane potential dyes are fluorescent probes that report changes in the electrical voltage across a cell membrane, providing a noninvasive way to monitor cellular excitability. These dyes redistribute, reorient, or alter their fluorescence in response to shifts in membrane potential, allowing optical signals to track depolarization and hyperpolarization over time. In biology, researchers use them to study neuronal signaling, muscle and cardiac activity, ion-channel function, and the effects of pharmacological treatments. Their ability to measure membrane dynamics across many cells simultaneously complements electrophysiological methods and supports investigations of cell communication, disease mechanisms, and drug responses.

The activity of single neurons from adult-aged mice can be studied by dissociating neurons from specific brain regions and using fluorescent membrane potential dye imaging. By testing responses to changes in glucose, this technique can be used to study the glucose sensitivity of adult ventromedial hypothalamic neurons.

Overview The relative difference in electrical charge, or voltage, between the inside and the outside of a cell membrane, is called the membrane potential. It is generated by differences in permeability of the membrane to various ions and the concentrations of these ions across the membrane. The Inside of a Neuron Is More Negative The membrane potential of a cell can be measured by inserting a microelectrode into a cell and comparing the charge to a reference electrode in the extracellular...
