Nkcc2 Symporter

The NKCC2 symporter is a membrane transport protein that moves sodium, potassium, and two chloride ions together across epithelial cells, making it essential for kidney function. Located on the apical membrane of cells in the thick ascending limb of the loop of Henle, NKCC2 uses the sodium electrochemical gradient to drive secondary active transport, while potassium recycling helps sustain its activity. Because this nephron segment is largely impermeable to water, NKCC2 builds the medullary osmotic gradient that enables later water reabsorption. Its activity supports urine concentration and electrolyte balance, and its inhibition by loop diuretics has important clinical applications.
