Hot Water Bath Digestion

Hot water bath digestion is a sample-preparation method in which controlled heating in a water bath helps break down a sample matrix and release substances of analytical interest. The sample is combined with an appropriate digestion reagent and heated indirectly, allowing water to transfer heat evenly while limiting temperature fluctuations, localized overheating, and vigorous boiling. This process can promote dissolution, hydrolysis, or decomposition of organic and inorganic components, depending on the reagents and sample composition. In chemistry, hot water bath digestion supports the preparation of biological, environmental, and material samples for subsequent analysis, improving analyte recovery, solution uniformity, and measurement reliability.

The present protocol describes the maceration and cleaning of cadaveric bone with a vacuum-sealed, hot water bath immersion technique. This is a low-cost, safe, and effective method to produce anatomical specimens for surgical planning and medical education as an alternative to three-dimensional (3D) printed models.

In response to the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, a laboratory protocol was developed to test the viral disinfection efficacy of hot water laundering of cloth face coverings, cotton scrubs, and denim pants. The Phi6 virus (bacteriophage) was used as the organism to test disinfection efficacy.

Here, we present a protocol to demonstrate the generation of ice when water is introduced to a cold bath of brine, as a secondary refrigerant, at a range of temperatures well below the freezing point of water. It can be used as an alternative way of producing ice for industry.
