Membrane Protein Localization

Membrane protein localization is the process by which proteins are positioned within or associated with cellular membranes, where their placement determines access to substrates, signaling partners, and transport pathways. It depends on features such as hydrophobic transmembrane segments, signal sequences, charged residues, and interactions with membrane lipids or targeting machinery that guide proteins to specific membrane regions and orientations. In chemistry and biochemistry, studying localization reveals how molecular structure controls protein folding, stability, and function. These principles support research on membrane transport, receptor signaling, drug binding, and disease-associated mislocalization, while methods such as fluorescence imaging, fractionation, and biochemical labeling help measure protein distribution.

Single-Molecule Localization Microscopy of Membrane Proteins using Single-Antibody Labeling

This protocol describes single-antibody labeling (SAL) to resolve the nanoscale spatial organization of plasma membrane proteins. By leveraging cumulative antibody-epitope interactions at the single-molecule level, membrane SAL (mSAL) maps local epitope distributions while simultaneously capturing antibody binding behavior in the native cellular environment.
