Thyroid Hormones

Thyroid hormones are endocrine signals produced by the thyroid gland that regulate metabolic activity, growth, development, and energy use throughout the body. The gland primarily releases thyroxine (T4), which is converted in peripheral tissues to the more active triiodothyronine (T3); T3 enters cells and binds nuclear receptors to alter gene transcription. In clinical practice, thyroid hormone measurements and regulatory markers help evaluate disorders such as hypothyroidism and hyperthyroidism, while replacement therapy, typically with levothyroxine, restores hormone action when production is inadequate. Understanding their synthesis, transport, metabolism, and cellular effects supports diagnosis, treatment selection, and monitoring of endocrine disease.

Hormones can be classified into three main types based on their chemical structures: steroids, peptides, and amines. Their actions are mediated by the specific receptors they bind to on target cells. Steroid hormones are derived from cholesterol and are lipophilic in nature. This allows them to readily traverse the lipid-rich cell membrane to bind to their intracellular receptors in the cytoplasm or nucleus. Once bound, the cytoplasmic hormone-receptor complex translocates to the nucleus.
