Linker-mediated Pcr

Linker-mediated PCR is a molecular genetics method that amplifies DNA adjacent to a known sequence, enabling researchers to identify previously unknown flanking regions. The process typically involves digesting genomic DNA with a restriction enzyme, ligating synthetic double-stranded linkers to the resulting fragments, and amplifying targets with one linker-specific primer and one primer directed toward the known sequence; nested PCR can improve specificity. This approach is useful for mapping transposon, viral, or engineered DNA integration sites, characterizing genomic rearrangements, and locating sequences in complex genomes. By connecting known and unknown DNA, linker-mediated PCR supports genome analysis and genetic engineering studies.

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.
